摘要
目的 运用粒细胞集落刺激因子诱导骨髓间充质干细胞(MSC)移植归巢到急性心梗区域,研究不同环境下MSC在心肌内的分化状态及程度。方法 选用SD雄性大鼠,体重200~250g,随机分为4组:急性心肌梗死(AMI)干绷胞移植+粒细胞集落刺激因子(G—CSF)动员组(20只)、AMI干细胞移植组(20只)以及单纯G-CSF组(20只)以及单纯对照组(10只)。于治疗4w后取材,检测心肌梗死区及其周围的血管密度值。免疫组化染色检测抗体增殖细胞核抗原(PCNA)、肌浆蛋白(myogenin)、肌球蛋白(myosin)、缝管连接蛋白43(Cx43)及结蛋白(desinmim)在移植骨髓间充质干细胞中的表达。并用共聚焦免疫荧光双标染色方法标测PCNA、myogenin。结果 胶体金标记的MSC经G-CSF诱导移植到心脏内在梗死部位与心肌生长为一体,颜色为黑色,存HE切片中移植细胞的胞浆呈现紫红色,极易识别。血管密度检测结果显示G-CSF+MSC移植组心肌梗死区血管密度明显高于其他各组。免疫组化染色检测抗体PCNA、myogenin、myosin、Cx43及desinmim在G-CSF+MSC移植组心肌梗死区中的表达明显。其他各组未见明显表达。结论 G-CSF能够明显动员MSC归巢,促进梗死区血管新生,在梗死区分化成新的心肌。
Objective To investigate the effects of bone marrow stem cells (MSC) mobilization with granulocyte colony stimulating factor (G-CSF) for the therapy of myocardial infarction (MI) in rats and to explore the differentiation of MSC. Methods 70 male SD rats weighted 200 - 250 g were randomly divided G-CSF + rMSC group ( n = 20), rMSC group ( n = 20), G-CSF group ( n = 20) and control group ( n = 10). The rats were killed 4 weeks later to detect blood vessel density in MI region. Expressions of proliferating cell nuclear antigen ( PCNA), myogenin, myosin and cannexin 43.4 gene symbol (Cx43) and desmin in MSC were determined by immunohistochemical assay, while PCNA and myogenin were detected by confucal immunofluorescence double labeled stain methods Results MSC labeled by colloidal gold induced by G-CSF were transplanted in MI region and infused into cardiac muscle, presenting black. HE slices noted fuchsia cellular plasma, being easily recognized. Neovascularization in G-CSF + rMSC group significantly increased than that of other groups. Expressions of PCNA, myogenin, myosin, Cx43 and desimin were positive in G-CSF + rMSC group, but not seen in other groups. Couclusions G-CSF could obviously motivate MSC homing and promote neovascularization and differentiate new cardiac muscle in MI region.
