食管鳞状细胞癌中PINCH蛋白的表达及意义 被引量:5

摘要 目的探讨PINCH蛋白在食管鳞状细胞癌中的表达及其临床病理意义。方法采用免疫组化ABC法检测10例正常食管粘膜组织和64例食管鳞状细胞癌组织中PINCH蛋白的表达,并分析其临床意义。结果PINCH蛋白在鳞癌组织相关间质中呈异质性表达,其阳性率为56.25%(36/64),明显高于正常食管粘膜组织(0/10)(P<0.05),差异有显著性,而且在肿瘤浸润边缘阳性更明显。PINCH蛋白表达与患者性别、年龄及鳞癌组织学分级无相关性;淋巴结转移组的鳞癌组织中PINCH蛋白阳性表达率为90%(18/20),未转移组鳞癌病例中阳性率为40.91%(18/44),二者相比差异有显著性(P<0.05)。结论PINCH蛋白在食管鳞癌中异常表达,可能利于肿瘤的浸润和转移。提示PINCH蛋白可作一个肿瘤相关间质的标志物来预测肿瘤浸润和淋巴结转移的能力。在肿瘤的预后判断上有重要意义,并可能成为与肿瘤间质相关的抗肿瘤治疗的一个新靶点。 Objective To investigate the expression and clinicopathological significance of PINCH protein in esophagus squamous cell carcinoma. Methods ABC immunohistochemical method was used to detect the expression of PINCH in paraffin block from 10 cases normal esophagus mucous tissue and 64 cases esophagus squamous cell carcinoma, and to study the significance of expression of PINCH protein. Results PINCH protein expression in tumor-associated stromal cells of esophagus squamous cell carcinoma was heterogeneous and its positive rate 56.25% (36/64) was higher than that of normal esophagus mocosa(P〈0.01). The statistically difference was significant, and the expression of PINCH at invasive edges was stronger areas, PINCH protein expression was not obviously correlated with patient's gender, age, and histological classification of squmous cell carcinoma. The positive rate of 90% (18/20) of the lymph node metastasis group was significantly higher than 40. 91% (18/44) of non-lymph node metastasis group (P〈0.05). Conclusion Abnormal expression of PINCH protein may play an important role in the tumor's invasion and metastasis, The results showed that PINCH protein may be a marker of tumor-associated stromal cells to predict the ability of invasion and lymph node metastasis of tumor. Moreover, the results also suggest a new target for tumor therapy.
