摘要
目的 观察基质金属蛋白酶-2(MMP-2)随动脉粥样硬化和血管钙化病理变化过程的量效变化,并探讨辛伐他汀对其影响。方法 健康雄性SD大鼠44只,体质量150~200g。随机分为5组:①对照组;②VitD3组;③高脂组;④高脂高钙组;⑤辛伐他汀组。应用免疫组化技术,对动脉粥样硬化大鼠主动脉MMP-2的表达进行测定。结果 高脂高钙组的MMP-2表达高于高脂组(P〈0.05)。辛伐他汀组的MMP-2表达低于高脂高钙组(P〈0.05)。辛伐他汀能抑制主动脉MMP-2的表达,并且随着MMP-2表达程度的降低,主动脉钙化程度也有所改善。结论 MMP-2表达与钙代谢及血管钙化密切相关。辛伐他汀减轻主动脉钙化的作用可能是通过影响MMP-2表达水平而实现的。
AIM To observe the changes of MMP-2 level during atherosclerosis and artery calcification, and to investigate the mechanism of simvastatin affecting the vessel calcium metabolism. METHODS Forty-four male SD white rats, with body weight of 150 -200 g, were divided into five groups at random:① baseline group( n = 8 ) : normal diet ; ② VitD3 group( n = 9 ) : normal diet + VitD3 ; ③ high-cholesterol diet group( n = 9 ) :high cholesterol diet ; ④ high-cholesterol-calcium diet group ( n = 9 ) .. high cholesterol and high calcium diet; ⑤Simvastatin group( n = 9 ) :high cholesterol and high calcium diet, with simvastatin 10 mg/( kg ·d) added. The content of MMP-2 in artery walls was determined by immunohistochemistry method. RESULTS The high-cholesterol-calcium diet group, compared with high-cholesterol diet group, had higher expression of MMP-2 (P 〈 0.05 ). While compared with high-cholesterol-calcium diet group, MMP-2 expression of the simvastatin group was lower (P 〈 0.05). CONCLUSION MMP-2 expression is correlated with calcium metabolism and artery calcification. Simvastatin can restrain the expression of MMP-2 in artery and therefore alleviate the artery calcification.
