摘要
将猪鲜精做体外获能处理,调整精子的浓度为每毫升5×107个精子,活力不低于0.5。受体母猪发情后注射HCG800IU,之后的24~30 h采用手术的方法按3种方式将精子注入生殖道内。缝合创口,常规饲养管理。结果表明:宫管接合部注入的受胎率为100%(5/5),平均产仔7.2头;输卵管伞部注入的受胎率为0(0/5);注入子宫角的受胎率为60%(3/5),平均产仔7.3头。该技术为充分挖掘优秀公猪个体的遗传资源和利用率、为精子介导法构建转基因猪提供了新的技术途径,同时为人类男性不孕症的治疗提供了可供参考的方法。
Swine fresh semen were eapacitated in vitro, and diluted to 5×10^7 sperms per mini-liter in order to maintain the activity of 0.5. 24-30 hours after treated with 800IU HCG, acceptors were done artificial insemination with 0.5mL of the sperm by operated with three manners including from link site of uterus-oviduct to oviduct (LUO), from umbrella to oviduct (UO), uterus angle(UA), and treated another side. Wounds were then sewed. According to common feeding, the results showed that the average of pregnant rate was 100 percent for LUO(5/5) and 7.2 piglets, zero for UO, 60 percent(3/5)and 7.3 piglets for UA. The test would supply a way for deeper dredging up gene resource and utilizing rate and construction of transgenic swine by sperm introducing. Meanwhile it would provide a reference method for some men with sterility disease.
