摘要
为了观察脊髓挤压伤后血-脊髓屏障(BSCB)的通透性变化,本研究选用体重180~200g成年雄性SD大鼠39只,随机分为对照组(n=3)和脊髓损伤组(n=6)。后者又分为伤后0、8、24、72h以及1周、2周等6组,每组6只,再分为A组、B组,每组各3只。脊髓损伤组A组的组织切片行H.E.染色,显微镜下观察脊髓损伤后的形态学变化。对照组和B组动物股静脉注射30g/L伊文思蓝,30min后,以温生理盐水及4%多聚甲醛灌注动物,取出脊髓,行冰冻切片,荧光显微镜下观察。结果观察到,脊髓挤压伤术后72h内,损伤区周围均可见伊文思蓝染色,至1周后损伤区周围未见伊文思蓝染色。本研究结果提示,脊髓损伤72h内可能为有效给药时间窗,1周后给药,药物对脊髓损伤的治疗作用将难以达到治疗目的。
To observe the changes of blood-spinal cord harrier(BSCB) permeability after compressive spinal, cord injury, thirty-nine male adult Sprague-Dawley rats( 180 - 200 g) were used in this study. There were one control group of 3 rats and six spinal cord injury groups of 36 rats which were assigned to subgroups of O h, 8 h, 24 h, 72 h, 1 w and 2 w (the time of post-injury). Spinal cord injury groups were further assigned to A and B groups, each of which contained 3 rats. H.E. staining was used in A group to observe morphologic changes of spinal cord after injury by microscope. Evans blue(30 g/L) was injected into rats of the control group and B group through the femoral vein, and then used warm saline and 4% paraformaldehyde to perfuse these rats 30 minutes later. Spinal cord was taken out and cryosectioned immediately for histological examination by a fluorescence mieroscope. We observed that the red fluorescence of Evans blue was seen around the lesion site within 72 h after spinal cord injury, but was not seen in spinal cord 1 week later after injury. The present results suggest that the effective time of drugs administration is possiblely within 72 h after spinal cord injury. It will be hard for drags to act on the injury if administrating drags one week later.
