摘要
采用UV-Vis光谱法研究了pH=7.25的溶液中甲基百里酚蓝(MTB)-铒(Er)(Ⅲ)配合物与鲱鱼精DNA的相互作用.MTB-Er(Ⅲ)-DNA的最大吸收峰为595 nm,比MTB蓝移5 nm,比MTB-Er(Ⅲ)蓝移9 nm,比MTB-DNA蓝移6 nm.DNA对MTB-Er(Ⅲ)有明显的增色效应.应用双波长物质的量比法和平衡透析物质的量比法对MTB-Er(Ⅲ)-DNA进行了研究,结果显示结合比nMTB∶nEr(Ⅲ)∶nDNA=13∶52∶1,MTB-Er(Ⅲ)配合物与DNA作用的表观结合常数K=3.28×106.
The interaction of Methylthymol Blue(MTB)-Er(Ⅲ) complexes with Herring Sperm DNA was inverstigated by UV-Vis spectrophotometric method in acidity buffer solution(pH = 7.25). The maximum absorption of formative Blue MTB-Er(Ⅲ)-DNA coordination compound was obtained at 595 nm. MTB-Er(Ⅲ)-DNA was found that the red shift of maximum absorption of the complex was 5 nm, 6 nm and 9 nm compared to the MTB, MTB-DNA and MTB-Er(Ⅲ) complex. DNA with stronger utilization of augment colour to MTB-Er(Ⅲ) According to balance percolation method and double wavelengh method, working out the combine ratio nMTB: mEr(Ⅲ):nDNA = 13:52: 1, combine constant of Methylth ymol Blue-Er(Ⅲ) complexe with DNA K = 3.28 × 10^6.
