摘要
了解TNF-α对人肝星状细胞表达MMPs和IL-8的影响.用TNF-α刺激培养的hHSC,用ELISA检测不同时间培养上清中的MMPs和IL-8的浓度.结果显示受TNF-α刺激后,MMP-1、MMP-2和IL-8水平为28h>24h>20h>4h-16h(P<0.05~0.001);MMP-3为28h>24h(P<0.01),24h>20h>4h-16h(P<0.05).TNF-α刺激后MMP-1和MMP-2产生较早,从8h起已高于对照组(P<0.05),而MMP-3的平均浓度,从20h起才高于对照组(P<0.01).提示TNF-α可刺激hHSC表达MMP-1、MMP-2、MMP-3和IL-8,其表达水平随刺激时间的延长而逐渐增加;未受TNF-α刺激的hHSC可表达一定量的MMPs,基本不表达IL-8.
To probe the effect of TNF -α on express of matrix metalloproteinases(MMPs) and IL- 8 in a culture medium of human hepatic stellate cells(hHSC), hHSC were cultured and stimulated by TNF- α. MMPs and IL- 8 in a culture were detected by ELISA at different time after TNF -α stimulation. The results shown that MMP - 1, MMP - 2 and IL - 8 level was in turn at 28h 〉24h 〉20h 〉4h-16h(P 〈0. 05 -0. 001 ) ,MMP - 3 at 28h 〉 24h( P 〈0. 01 ) 〉 20h 〉 4h-16h( P 〈 0.05 ). Average level of MMP - 1 and MMP - 2 in TNF - α group was higher than control after a 8 hours stimulation respectively ( P 〈 0.05 ), and average level of MMP - 3 was not significantly higher than control till at 20h ( P 〈 0. 01 ). The present data indicated that TNF - α could stimulate hHSC to express MMP - 1, MMP - 2, MMP - 3 and IL - 8, and hHSC not stimulated by TNF- α may express a little MMPs but no IL- 8.
