EB病毒转化的系统性红斑狼疮家系患者B淋巴细胞株凋亡相关基因的表达研究 被引量:1

摘要 目的检测EB病毒转化的系统性红斑狼疮(SLE)家系患者B淋巴细胞株凋亡相关基因的表达情况,探索凋亡在SLE发病中的作用。方法收集到4个SLE家系,共20个成员,其中SLE患者8人,用EB病毒对其家系成员外周血淋巴细胞进行了转化建株,流式细胞仪等鉴定其表型;采用荧光标记半定量逆转录聚合酶链反应(RT-PCR)方法检测了凋亡相关基因sFas、Fas和Bc l-2的表达。结果成功建立了SLE家系“永生型”淋巴细胞株,建株细胞表达CD20,而不表达CD3,且具有抗体分泌功能,为B淋巴细胞;凋亡相关基因sFas、Fas和Bc l-2 mRNA的表达在SLE家系的患者和非患者成员之间均未见显著性差异(分别为1.347±0.238 vs 1.222±0.215,8.929±2.572 vs 7.454±1.367和5.296±1.422 vs 5.534±1.495,P>0.05)。结论凋亡相关基因sFas、Fas和Bc l-2等可能不是SLE的易感基因,其在SLE的发病中有待于进一步研究。 Objective To study the expressions of the apoptosis - related genes of EB virus - transformed B cell strains in SLE family patients to explore their roles in the pathogenesis of SLE. Methods The PBMLs of 4 SLE families ( totally 20 members, 8 SLE patients) were transformed with EB virus and cultured. Their phenotypes were identified with cytometric assay, mRNA expressions of sFas,Fas and Bcl -2 genes were then measured with fluorescence - labeled semi - quantitative RT - PCR. Results B cell strains of SLE families were successfully set up, which express CD20, not express CD3 and can secret immunoglobulin, mRNA expressions of sFas, Fas and Bcl - 2 genes in the patients and other members of SLE family were I. 347 ± 0. 238 vs 1,222±0.215, 8. 929±2. 572 vs 7. 454±1. 367 and 5. 296±1. 422 vs 5. 534±1. 495 resectively and there was no statistical difference( P〉0.05 ). Conclusions Apoptosis - related genes are not critical to the pathogenesis of SLE.
