生长抑素类似物奥曲肽中枢抑制大鼠胃酸分泌的机制 被引量:2

摘要 目的:观察第三脑室预先注射纳洛酮或急性膈下迷走神经切断术对第三脑室注射生长抑素类似物奥曲肽抑制大鼠胃酸分泌的影响。方法:实验用♂Wistar大鼠,第三脑室埋管一周后,进行急性胃灌流实验,在皮下注射五肽胃泌素的基础上,第三脑室注射生理盐水(A组)、第三脑室注射奥曲肽0.05μg(B组)、第三脑室注射纳洛酮2.5μg+奥曲肽0.05μg(C组)、急性膈下迷走神经切断术+第三脑室注射生理盐水(D组)、或急性膈下迷走神经切断术+第三脑室注射奥曲肽0.05μg(E组),分别测定1h总酸排出量,并与脑室注药前诱导的1h总酸排出量进行比较,计算其变化百分率。结果:1h总酸排出量变化率分别为:A组4.60%、B组-20.35%、C组-18.06%、D组5.01%、E组-21.59%。B组或C组与A组比较P<0.01;E组与D组比较P<0.01;C组或E组与B组比较,组间差异无显著性(均P>0.05)。结论:(1)奥曲肽抑制胃酸分泌的作用在中枢可能不需内源性吗啡样物质或其受体介导;(2)奥曲肽抑制胃酸分泌的效应在外周是通过迷走外途径介导的。 OBJECTIVE To investigate the effect of preceding naloxone rejection into the third cerebroventricle(icv) or acute subdiaphragmatic vagotomy on the gastric acid secretion inhibited by the somatostatin analogue octreotide given icv. METHODS The third ventricles were cannulated in male Wistar rats. One week later,acute gastric lumen perfusion was carried out. On the basis of subcutaneous injection of pentagastrin, icy physiological saline (Group A), icy octreotide 0. 05 μg (Group B), icv naloxone 2. 5 μg octreotide 0. 05 μg (Group C), acute subdiaphragmatic vagotomy +icv physiological saline (Group D), or acute subdiaphragrnatic vagotomy +icv octreotide 0. 05 μg (Group E) were conducted. Before and after icv injection, total acid output (TAO) of lh was determined and compared. The experimental data were expressed in change rate (%) of TAO. RESULTS The change rates (%) of TAO were 4. 60% in Group A, - 20. 35% in group B, - 18. 06% in Group C,5.01 % in Group D and - 21.59% in Group E, respectively. Comparison of Group 13 or C versus Group A showed (P〈0. 01) ,Group E versus Group D (P〈0. 01 ). Whereas, the differences between Group C and Group B,Group E and Group B were statistically insignificant (P〈 0. 05). CONCLUSION The results indicate: ( 1 )The central inhibition of gastric acid secretion by octreotide may not he mediated by the endogenous opiate substance or its receptor; (2)The peripheral pathway for icv octreotide to suppress gastric acid secretion is via extra-vagus route.
