摘要
目的:观察加味止嗽散(MZ)、MZ脂溶性部位群(MZC)和水溶性部位群(MZS)对过敏性哮喘豚鼠血液和支气管肺泡灌洗液(BALF)中嗜酸粒细胞(EOS)、内皮素-1(ET-1)、一氧化氮(NO)含量及对肺组织结构的影响。方法:采用Wright染色法计血液和BALF中的EOS。采用放射免疫法和硝酸还原酶法分别测定哮喘豚鼠血清和BALF中ET-1、NO的含量。光镜和电镜下观察豚鼠肺组织的结构变化。结果:哮喘模型组豚鼠体内的EOS、ET-1、NO的量明显高于正常对照组(P<0.01),肺组织病理学和超微结构发生明显改变。与哮喘模型组比较,MZ和MZC治疗组能降低豚鼠血液和BALF中EOS、ET-1、NO的含量(P<0.01或P<0.01)和减轻肺组织病理改变,MZS组则未见有明显的作用。结论:MZ中具有平喘作用的物质大多数集中在MZC中,其平喘作用机制可能与其能减少哮喘豚鼠体内EOS数量、恢复ET-1、NO平衡和减轻肺组织结构损伤有关。
Objective : To study the effects of different fractions of Modified Zhisousan ( MZ, MZC, MZS) on the contents of nitric oxide (NO), endothelin-1 (ET-1) and eosinophilia (EOS) in the allergic asthma guinea pigs and observe the pathology changes of lung tissue, Methods: The number of EOS in the blood and bronchialveolar lavage fluid (BALF) was counted by Wright staining. The contents of ET-I and No in serum and BALF were analyzed by RIA and nitric acid reductase method. The guinea pig lungs were ob- served under the optical and electron microscope. Results: The number of EOS and the contents of ET-1 and NO in model group were higher than those in control group ( P 〈 0.01 ). The pathological changes of lung were obvious. The number of EOS and the contents of ET-1 and NO were descended remarkably ( P 〈 0.05 or P 〈 0. 01 ) and the pathological changes in the lung tissue were lightened obviously in MZ and MZC groups, but MZS group had no such effects. Conclusion : MZC is the effective part of MZ and the anti-asthmatic mechanisms ware related to its significant reduction in contents of ET-1, NO, EOS and the damage of lung tissue possibly.
