摘要 目的:克隆人成牙本质细胞L型钙离子通道α1亚基D亚型的特异性基因。方法:采用RT-PCR方法,从人成牙本质样细胞系中克隆人成牙本质细胞L型钙离子通道α1亚基的特异性基因片段,将其亚克隆入pUC18载体进行序列测定。结果:从人成牙本质样细胞系中获得900bp的特异性片段。序列分析表明,与Gen-Bank登陆的人L型钙离子通道α1亚基D亚型的特异性基因序列完全一致。结论:成功地从人成牙本质样细胞系中克隆到人成牙本质细胞L型钙离子通道α1亚基D亚型的特异性基因序列。 Objective:To clone L type calcium channel α1 D subunit specific gene of human odontoblasts. Methods: L type calcium channel α1 D subunit specific gene of human odontoblasts was cloned from human odontoblast-like cell line by RT-PCR, target fragment was inserted into pUC18 vector and sequenced. Results:A 900 bp fragment was obtained from human odontoblast-like cell line. The sequence analysis showed that it was 99.9% homologous to human L type calcium channel α1 D subunit specific gene reported by Genebank. Conclusion: The specific sequence of L type calcium channel α1 D subunit of human odontoblast is successfully cloned from human odontoblast-like cell line.
