摘要 目的:探讨肝癌大鼠免疫系统的昼夜节律变化。方法:肝癌大鼠模型在12:12明暗交替环境中生长四周后,每4h采集一次血液标本,检测免疫指标。结果:①正常大鼠的免疫系统存在昼夜节律。②肝癌大鼠的免疫系统节律发生紊乱。结论:肝癌大鼠的免疫系统昼夜节律已发生紊乱,提示肝癌的生长和免疫系统的昼夜节律之间存在着相关关系,为时间药理学和时间免疫学等基础研究及临床择时治疗提供理论依据。 Objective :To explore the circadian rhythm of immune system in liver cancer rats. Methods : The Wistar rats which were adapted to synchronized with an alternation of 12 h of light(L) and 12 h of darkness(D) (LD12: 12) were planted cancer piece into liver. After 4 weeks, the blood samples as well as the normal were collected 6 times a day(8:00,12100,16:00,20:00,24:00,4:00 ). The number of WBC, percentage of neutrophil and lymphocyte were detected by cell-counter. Level of interleukin 2(IL-2) and interleukin 6(IL-6) were measured by using ELISA technique. Results: The immune system of liver cancer rats demonstrated difference from the normal group. The number of WBC in circulation exposed similar rhythm with the normal and that got to its peak value at the hour of 12:00-16:00 and followed by a trough at 8:00. The whole rhythm showed as ladle-like, but the phase weakened. The acrophase in percentage of neutrophil and lymphocyte showed the reverse type and single peak. The aerophase of the level of IL-2 had a tendency to go forward than the normal group and that got to peak val ue at 12:00 hour but lowered at 8:00. No diurnal rhythm of IL-6 was observed. While rhythm of IL-6 showed a variation like "W", the peak value was seen at the hour of 4 : 00 and 16 : 00 and then followed by a lowest point at 12100 hour in normal group. Conclusion: This finding suggests that immune system in normal rats exists circadian rhythm and alteration in rats with liver cancer. The development of liver cancer may correlate with the rhythm of immune system, which will provide a theo- retical source for researching chrono-pharmaeology ,cbrono-immunology and chrono-therapy.
