摘要
目的:在体外分离纯化VX2肿瘤细胞,观察其生物学特性.方法:采用组织块法和消化法结合对兔VX2肿瘤进行原代培养,体外传代观察,传代40次并对培养细胞进行形态学观察、细胞周期检查、核型分析、兔及裸鼠移植.结果:纯化的兔VX2肿瘤细胞形态一致,呈圆形、多角形的上皮细胞,体积小,核浆比倒置,体外连续培养8mo,传代50次以上,细胞倍增时间为26.4h,细胞周期测定G1期为74.61%,G2期为7.78%,S期为17.6%.染色体为亚三倍体核型,众数为60条.高细胞浓度可保证同种移植成瘤率,裸鼠移植可成瘤,无支原体污染.结论:兔VX2肿瘤细胞系来源于Shope病毒致乳头瘤恶变形成的鳞状上皮细胞恶性肿瘤,目前已经纯化,可应用于进一步肿瘤实验研究.
AIM: To separate and purify the rabbit VX2 carcinoma cell line in vitro and to characterize its biological features. METHODS: VX2 tumor fragments were isolated and cultured with explant culture techniques and enzymatic digestion in vitro. After 40 passages, the cell morphology, cell cycle, karyotype and tumorigenecity in rabbits and nude mice were investigated. RESULTS: The newly established cell line VX2 was maintained in continuous culture over 50 passages for 8 months. Morphologically, VX2 ceils were uniform, round and polygonal epithelial ceils. The ceils were small but had a high nucleocytoplasmic ratio. The cell cycle analysis showed 74. 61% in G1 phase, 7.78% in G2 phase and 17.6% in S phase. The population doubling time was 26. 4 h. The chromosomal analysis showed a hypo-triploid with a median chromosome number of 60. The tumorigenecity in rabbits and nude mice will be ensured under a high VX2 cell inoculating concentration. CONCLUSION: The VX2 is a squamous-cell carcinoma resulting from malignant change in a papilloma induced with the Shope vires in rabbits. It has already been purified in vitro, and can be applied to further researches.
