摘要
目的对细粒棘球蚴重组抗原EF-1的免疫原性进行初步鉴定,为重组疫苗研制的后续工作提供依据。方法用重组目的蛋白及融合表达蛋白作为抗原,分别免疫昆明小鼠获得抗血清,通过蛋白免疫印迹试验 (Western blot)和酶联免疫吸附试验(ELISA)检测血清中产生的特异性抗体成分及其程度。结果重组目的蛋白EF-1和融合蛋白GST/EF-1免疫小鼠后均诱发产生了特异性抗体,实验组血清与对照组血清抗体含量差异有统计学意义(P<0.05);目的蛋白和融合蛋白免疫两组小鼠所得血清的抗体含量差异没有统计学意义。结论目的蛋白EF-1和融合蛋白GST/EF-1都具有一定的免疫原性。
Objective To evaluate the immunogenicity of recombinant antigen EF- 1 of echinococcusis granulosus, and to pave a base for the following study of recombinant vaccine. Methods The recombinated interest protein and fusion protein as antigens were injected respectively into Kunming mice in order to induce the specific antibodies. Then the composition and concentration of the specific antibodies in serum were detected by Western blot and ELISA. Results The specific antibodies of recombinant antigen EF - 1 and fusion protein GST/EF - 1 were induced and detected, these antibodies were significantly different between subjects and control, but no difference between the antibody concentrations induced by those two antigens. Conclusion The interest protein EF- 1 and fusion protein GST/EF- 1 have certain immunogenicity.
