摘要
目的观察早期和晚期吸入糖皮质激素对哮喘大鼠气道炎症和气道重构的影响。方法32只Wistar大鼠随机分为4组:A对照组8只,B卵蛋白(OVA)致哮喘组8只,C早期激素治疗组8只,D晚期激素治疗组8只。各组大鼠肺组织切片HE、胶原染色、免疫组化NGF、TGF-β1染色,借助计算机图像分析软件测量各项指标。结果B组气道壁炎性细胞计数、气道平滑肌面积、气道内壁面积、胶原沉积面积及NGF、TGF-β1表达均明显高于A组,两组比较差异有统计学意义(P<0.01),C组各项指标均明显低于B组,差异有统计学意义(P<0.01、P<0.05),C组与A组比较各项均有升高,差异有统计学意义(P<0.01、P<0.05),D组仅气道壁炎性细胞计数、NGF表达低于B组,差异有统计学意义(P<0.05),C组各项与D组比较差异均有统计学意义(P<0.01、P<0.05)。结论早期吸入激素可明显抑制气道炎症和气道重构,晚期吸入仅能部分抑制炎症反应,不能逆转已经形成的气道重构。
Objective To investigate the effects of inhaled glucocorticoid in early phase and late phase on the airway inflammation and remodeling in asthmatic rots.Methods Thirty-two Wistar rats were randomly divided into 4 groups:group A( n = 8,control with saline) group B( n = 8, asthma with OVA) ,group C( n = 8, before OVA challenging treated with glucocorticoid in early phase), and group D( n = 8, after OVA Challenging repeatedly for 4 weeks treated with glucocorticoid in late phase). The pathologic alteration of the bronchi and lung tissue was observed by HE staining. Expression of NGF and TGF-β1 were detected by immunohistochemistry, collagen deposition by Masson staining. The Morphological parameters were measured by computer image analysis software.Results Group B,the inflammatory cell counts around bronchus,airway smooth muscle hypertrophy, airway inner wall area, the collagen deposition of reticular basement, were significant higher than those of group A( P 〈 0.01). Group C were significantly lower than group B ( P 〈 0.01, P 〈 0.05) .There were apparent difference between group C and group A( P 〈 0.01, P 〈 0.05). Group D, the inflammatory cell counts around bronchus, the expression of NGF significantly reduced as compared with group B ( P 〈 0.05). There were apparent difference between group C and group D. ( P 〈 0.01, P 〈 0.05). Conclusion Inhaled glucocortieoid could apparently inhibit airway inflammation and remodeling in early phase, but could partially inhibit airway inflammation and not reverse the airway remodeling in late phase.
