IPGphor^(TM)等电聚焦加样方法的改进与电泳结果的预测

摘要 固相pH梯度干胶条等电聚焦电泳技术因其分辨率高而成为当今双向电泳分离蛋白质的首选技术,但其实验步骤多难度大,要求试剂配制精确,各项操作无误。通过改变蛋白质加样方法,测定固相pH梯度干胶条IEF电泳过程中的前期水化干胶条时初始电流与终电压的内在规律,发现在不同的样品和不同的设定电压条件下初始电流的变化与IEF电泳终电压结果存在着高度的相关性,水化干胶条时的初始电流与终电压亦存在着高度的相关性。将IEF电泳的水化初始电流值(1μA)作为蛋白质等电聚焦判断标准,指导实验。其方法实用简便,可提高工作效率。 IPGphor^TM IEF is using as the first technology for protein separation nowadays for its high efficiency, but the operation is much trublesome, needing to prepare resolution precisely, without any error operation in this course. Through the change of protein-adding methods during the operation of IPGphor^TM IEF, we find out some basic principles relating to initial electric currency and final electric voltage at precious of IPGphor^TM hydration. That is highly relationship exists between initial electric currency and final voltage under different designed voltage for different samples. The same relationship also exists when IPGphor^TM is hydrating. Using the initial electric currency (1μA) as judgment standard and guiding experiment when IEF of protein. This method is practical simple and high efficiency.
