茶色素对吗啡依赖小鼠和大鼠的戒毒作用 被引量:4

摘要 目的:观察茶叶中提取物茶色素对吗啡依赖小鼠和大鼠催促戒断症状的抑制作用并分析其可能的作用机制。方法:实验于2003-03/2004-03在大连医科大学药理教研室完成。选择昆明种小鼠110只;SD大鼠40只,将小鼠随机分为正常对照组(n=20)、吗啡依赖组(n=20)、茶色素高、中、低剂量组(n=20),左旋咪唑阳性对照组(n=10)。将所有大鼠随机分为正常对照组、吗啡依赖组、茶色素高、低剂量组,每组10只。所有动物均采用皮下注射吗啡的方法建立小鼠及大鼠吗啡依赖模型。采用催促戒断治疗试验测定10min内小鼠及大鼠出现的跳跃反应次数及潜伏期,比较跳跃反应前后1h的体质量变化,同时进行免疫功能测定,超氧化物歧化酶活力和丙二醛含量的测定。结果:各实验组小鼠及大鼠全部进入结果分析。①催促戒断治疗试验结果:与小鼠吗啡依赖组比较,高剂量组的小鼠跳跃次数明显减少(P<0.05),高、中剂量组的小鼠跳跃反应潜伏时间均明显延长(P<0.05)。跳跃反应后,高剂量组体质量明显下降(P<0.05),与大鼠吗啡依赖组比较,大鼠茶色素高、低剂量组的戒断症状分值均明显降低,高剂量组的体质量明显下降。②吗啡依赖小鼠免疫功能的测定结果:与吗啡依赖组比较,茶色素高、中、低剂量组及左旋咪唑组的血清半数溶血值均有提高,并具有显著性差异(P<0.05),茶色素高、中剂量组及左旋咪唑组能显著提高小鼠对二硝基氟苯所致足掌肿胀值(P<0.01~0.05),茶色素高剂量组及左旋咪唑组的胸腺指数提高(P<0.05)。③超氧化物歧化酶活力和丙二醛含量的测定结果:与正常对照组比较,吗啡依赖组小鼠超氧化物歧化酶活力明显降低,丙二醛含量明显升高(P<0.05)。与吗啡依赖组相比,茶色素高剂量组能显著提高小鼠的超氧化物歧化酶活力,中剂量组虽也提高了小鼠血清中超氧化物歧化酶活力但差异无显著性, AIM: To investigate inhibitory effect of tea pigment on withdrawal syndrome of morphine-dependent mice and rats and analyze its possible mechanism. METHODS: The experiment was completed in the Pharmacological Department of Dalian Medical University from March 2003 to March 2004. Totally 110 Kunming mice were divided randomly into normal control group (n=20), morphine-dependent group (n=20), high, middle and low dosage of tea pigment groups (n=20) and levamisole positive control group (n=10);40 SD rats were divided randomly normal control group, morphine- dependent group, high and low dosage of tea pigment groups with 10 in each group, Morphine-dependent models of mice and rats were established with subcutaneous injection, respectively. Jumping times and latency of mice were measured within 10 minutes, and changes of body mass were compared 1 hour before and after jumping response. Immunological functions, activity of superoxide dismutase (SOD) and content of malondialdehyde (MDA) of mice were assayed, respectively. RESULTS: All mice and rats entered the final analysis.① Results of withdrawal test: Jumping times of mice in high dosage group was less than those in morphine-dependent group (P 〈 0.05), and latency of jumping response was longer in high and middle dosage groups than that in morphine-dependent group (P 〈 0.05). After jumping response, body mass of mice in high dosage group was decreased remarkably (P 〈 0.05), and as compared with rats in morphine-dependent group, scores of withdrawal symptom in high and low dosage groups were decreased obviously, and also body mass in high dosage group. ② Results of immunological functions of mice in morphine-dependent group: As compared with those in morphine-dependent group, serum semi-hemolysis value in three dosage groups and levamisole group was increased, and there was significantly different (P 〈 0.05); swelling value of feet induced by dinitrofluorobenzene was increased in three dosage groups and
