摘要
目的研究安君宁对雄性SD大鼠吗啡依赖相关脑区前脑啡肽原表达的影响。方法30只体质量180~220g的雄性SD大鼠,设对照并建立吗啡依赖模型后随机均衡分为5组:淀粉正常对照组,吗啡戒断并安君宁治疗12d、30d组,吗啡戒断并淀粉治疗12d、30d组。吗啡依赖模型的建立采用剂量递增10日法(5mg·kg-1·次-1~50mg·kg-1·次-1,2次/d,腹腔注射)。安君宁干预方法:灌胃1g·kg-1·次-1,2次/d;对照组灌等量的淀粉。各组大鼠按预定时间麻醉、灌注、留取脑组织。采用原位杂交技术测定腹侧被盖区(VTA)、伏隔核(NAs)、前额皮质(PFC)前脑啡肽原(PENK)mRNA水平。结果安君宁干预组与干预对照组相比,VTA、NAs、PFC中PENKmRNA水平显著升高(P<0.05)。吗啡戒断12d,安君宁干预组VTA、NAs、PFC中PENKmRNA水平分别为(14.74±4.81)×10-2、(16.09±4.94)×10-2、(20.47±7.03)×10-2,而干预对照组分别为(9.74±0.87)×10-2、(5.94±0.91)×10-2、(10.87±3.96)×10-2;戒断30d,安君宁干预组分别为(17.46±8.27)×10-2、(25.73±8.24)×10-2、(26.58±6.29)×10-2,而干预对照组分别为(12.36±6.95)×10-2、(10.31±0.78)×10-2、(19.78±7.08)×102。结论安君宁能促进吗啡依赖大鼠依赖相关脑区前脑啡肽原表达的恢复。
Objective To explore the impacts of Anjunning on transcription of pre-proenkephalin genes in some brain region of morphine dependent Spragne-Dawley rats. Methods Thirty male Sprague-Dawley rats weighing 180 -220g were assigned randomly and averagely into one normal group and four abstinent treatment group : (1) Normal group, injected with saline, (2) P12E group, abstinent after the establishment of morphine dependence model and treated with Anjunning for 12 days; (3) P12EC group, abstinent and treated with starch for 12 days; (4) P30E group, abstinent and treated with Anjunning for 30 days; (5) P30EC group, abstinent and treated with starch for 30 days. The morphine dependence model was established by injecting morphine intraperitoneally in increasing dose regime (5 -50mg/kg, twice a day) for 10 days. Anjunning were administrated twice a day in dosage of 1g/kg after termination of morphine injection, accordingly starch in the same volume as Anjunning was administrated to control group by same route. After rats were anesthetized and infused with formaldehydum polymerisatum, then sacrificed, the brain were removed and cut into sections. The levels of mRNA of pre-proenkephalin in ventral tegmental area, nucleus accumbens septi, prefrontal cortex were determined by in situ hybridization. Results Compared with control group, the levels of mRNA of Pre-proenkephalin in ventral tegmental area, nucleus accumbens septi, prefrontal cortex also increased. ( P 〈 0.05 ). Conclusion Anjunning can improve the recovery of the functional disturbance of pre-proenkephalin for the morphine dependent rats, which may be the mechanism anjunning in alleviating the heroin protracted abstinence symptoms, especially craving, which had been observed in our clinical research.
