骨折损伤后不同时相骨髓间充质干细胞表面生长因子受体表达的变化

摘要 目的:观察骨折损伤后不同时相骨髓间充质干细胞表面生长因子受体表达的变化,分析其对骨折损伤后组织修复的作用。方法:实验于2002-02/11在解放军第三军医大学预防医学院复合伤研究所,创伤、烧伤与复合伤国家重点实验室完成。①建立成兔桡骨骨折模型。②创前(0d)及创后1,5,10,15,20d分别采取骨髓有核细胞,纯化间充质干细胞。③成骨分化培养基诱导培养间充质干细胞。④通过Western-blot法检测碱性成纤维细胞因子受体Bek及骨形态发生蛋白受体-Ⅱ在间充质干细胞上的表达。结果:①碱性成纤维细胞因子受体Bek表达:创后1d,碱性成纤维细胞因子受体Bek表达与0d比较无明显差别(P>0.05);创后5d,Bek表达与0d相比显著增高(P<0.05);创后10d与5d比较无明显差别(P>0.05),但与0d相比,其表达显著增高(P<0.05);创后15d,20d的Bek表达较0,5d及10d均明显增高(P<0.01)。②骨形态发生蛋白受体-Ⅱ表达:创后1d,5d,骨形态发生蛋白受体-Ⅱ表达与0d比较无明显差别(P>0.05);创后10d,15d其表达与0d比较显著增高(P<0.05);创后20d其表达与0,1,5,10,15d相比,明显增高(P<0.01)。结论:创伤后体内骨髓间充质干细胞成骨分化过程中其碱性成纤维细胞因子受体Bek及骨形态发生蛋白受体-Ⅱ的表达逐渐增高。 AIM: To investigate the expression of growth factor receptors on the surface of bone marrow-derived mesenchymal stem cells at different phases after bone trauma and analyze its effects on tissue repair after bone trauma. METHODS: The experiment was carried out at State Key Laboratory of Trauma, Burns and Combined Injury, Institute of Combined Injury, College of Preventive Medicine, Third Military Medical University of Chinese PLA from February to November 2002. ①Establishing the model of fracture of radius of adult rabbit, ②Harvesting bone-marrow karyotes before trauma and 1 day, 5 days, 10 days, 15 days, 20 days after trauma and purifying mesenchymal stem cells, ③Cuhuring with inducing osteogenesis culture medium④Detecting the expression of basic fibroblast growth factor (bFGF) receptor Bek and bone morphogenic protein receptor-Ⅱ (BMPR-Ⅱ ) on mesenchymal stem cells with Western-blot method. RESULTS:①The expression of basic fibroblast growth factor receptor Bek had no significant difference between two groups on the 1^st day and 0^th day (P 〉 0.05);the expression of Bek increased on the 5^th day after trauma and was significantly higher than that before trauma (P 〈 0.05); there was no obvious difference between the 10^th day and 5^th da^th after trauma(P 〉 0.05) ,but it was significantly higher than that on the 0^th day (P 〈 0.05) ;The expression of Bek on the 15^th day and 20^th day were both very significantly higher than that before trauma, on the 5^th day and 10^th day after trauma(P 〈 0.01 ). ②The expression of bone morphogenic protein receptor-Ⅱ (BMPR-Ⅱ ) had no significant difference between the two groups on the 1^th day and before trauma (P 〉 0.05);it increased on the 5^th day after trauma but there was no obvious difference compared with that before trauma (P 〉 0.05); The expression of BMP- Ⅱ increased on the 10^th day and 15^th day after trauma and was significantly higher than that before trauma (P 〈 0.05);The
