摘要 目的探讨高渗盐液预处理对大鼠肾缺血再灌注损伤后肾组织细胞间黏附分子-1的表达的影响。方法56只SD大鼠随机分为假手术对照组(S组)、缺血再灌注组(IR组)和高渗盐液处理组(H组)。肾缺血再灌注模型的建立:左侧肾蒂夹闭45min后松开,分别于再灌注4、6、12h计算左肾系数,并分别用免疫组化和逆转录-聚合酶联反应(RT-PCR)测定肾组织的细胞间黏附分子-1的蛋白以及mRNA的表达。结果H组左肾系数和肾组织ICAM-1蛋白及mRNA的表达明显低于IR组。结论高渗盐液预处理能下调缺血再灌注后肾脏ICAM-1的蛋白和mRNA表达,对肾缺血再灌注损伤有保护作用。 Objective To explore the effects of hypertonic saline preconditioning on the expression of intercellular adhesion mol ecules-1 (ICAM -1 )after renal isehemia-reperfusion injury in rats. Methods Fifty-six SD rats were divided randomly into the sham operative control group (S group), renal ischemia-reperfusion injury group fIR group) and hypertonie saline-treated group (H group) . The model of renal isehemia-reperfusion in rats was established by clamping the left renal pedicel in situ for 45min before reperfusion of blood 4h, 6h or 12h. The left renal coefficeince was calculated, the expression of ICAM-1 protein and mRNA was measured by immunohistochemieal and reverse transcription polymerlymerase chain reaction (RT-PCR) individually. Results The left renal coeffieeinee,the expression of ICAM- 1 protein and mRNA were lower in H group compared with IR group. Conclusion Hypertonie saline preconditioning can protect the renal tissue after renal ischemia-reperfusion by down-regulating the ICAM-1 protein and mRNA in rats.
