摘要
目的观察不同水平+Cz作用后,大鼠血清和支气管肺泡灌洗液(BALF)中炎性细胞因子含量、氧化和抗氧化物含量和蛋白总量的变化,以及BALF中白细胞(WBC)总数和分类的变化, 了解持续性高加速度引起的肺损伤的作用机制。方法雄性Wistar大鼠20只,随机分为对照组、 +5 G组、+10 G组和+15 G组(n=5)。采用梯形+Gz曲线,+5 G和+10 G各暴露90 s,+15 G 暴露30 s,并间隔30 min重复3次。于暴露后4 h取大鼠腹腔静脉血、BALF及肺组织,测定血清和 BALF中自介素-1β(IL-1β)、白介素-8(IL-8)和肿瘤坏死因子-α(TNF-α),肺组织匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)和总蛋白(TP),BALF中WBC及分类。结果 +Gz作用后,血清和 BALF中IL-1和TNF-α、肺组织匀浆中MDA和TP含量、BALF中WBC和多核细胞均明显增加,且随着+Gz水平的升高而升高,而SOD含量减少。但IL-8变化不明显。结论 +Gz暴露后,大鼠肺组织局部炎性细胞和炎性细胞因子增加、氧化抗氧化失去平衡,提示炎症细胞和炎症细胞因子在持续性高加速度引起的大鼠肺脏病变中起重要作用。
Objective To observe the changes of the concentrations of cytokines, oxidantantioxidant and total protein (TP) in rats' serum, bronchoalveolar lavage fluid (BALF) and lung homogenate, as well as WBC count and differential count in BALF, following different levels of high +Gz exposures, so as to explore the mechanism of lung injury induced by sustained high +Gz. Methods Twenty male healthy Wistar rats were randomly divided into 4 groups (n= 5 ) .. control group, +5 G group, + 10 G group and + 15 G group. Profiles of the centrifuge +Gz exposures were trapezoidal, +5 G and +10 G lasted for 90 s, +15 G group for 30 s. The +G, profile repeated for 3 times with 30 rain interval in each group. The blood sample, BALF and lung tissue were collected 4 h after the last centrifuge run. The concentrations of IL-1β, IL-8 and TNF-α in the serum and BALF, SOD, MDA and TP in the lung homogenate as well as WBC counts in the BALF were examined. Results The concentrations of IL-1β and TNF-α in serum and BALF, MDA and TP in the lung homogenate and WBC count and the rate of polyeyte in the BALF of high +Gz exposure groups were significantly higher than those of controls, along with the enhancement of +Gz level these changes become more prominant. The concentrations of SOD become significantly lower but IL-8 remained unchanged after high + Gz exposures. Conclusions The results indicate that the increase of inflammatory cells and cytokines may play important roles in the pathogenesis of lung injury induced by sustained high + Gz exposures.
