摘要
目的:采用酶组织化学法观察缺血预处理对大鼠骨骼肌缺血再灌注损伤是否有保护作用。方法:实验于2005-07/10在锦州医学院药理实验室完成。选取健康清洁级1月龄SD大鼠24只,随机分为3组:假手术对照组、缺血再灌注组、经典缺血预处理组,8只/组。①缺血再灌注组和经典缺血预处理组大鼠建立骨骼肌缺血/再灌注模型。大鼠腹腔麻醉后沿股鞘切开皮肤,分离股动静脉,制成切断患肢皮肤、肌肉和神经,仅保留股动静脉,用微血管夹阻断股动脉血供,制成右后肢缺血模型。②缺血再灌注组大鼠造模后缺血60min,再灌注20min。经典缺血预处理组大鼠用无创伤血管夹阻断股动静脉,给予10min的短暂缺血及10min的再灌注,3次循环后再次阻断股动静脉,缝合左大腿前方皮肤切口,缺血60min,剪开左大腿前方切口取出无创伤血管夹,观察血流恢复无血栓形成后,在动脉血流恢复的情况下可观察到患肢鼠爪由暗紫色转变为粉红色,再灌注20min。假手术对照组大鼠只穿线不结扎,股动脉旷置50min。③通过光镜观察各组骨骼肌结构的变化,同时以黄嘌呤氧化酶法测定各组大鼠骨骼肌组织的超氧化物歧化酶活性,硫代巴妥酸法测定丙二醛含量,酶组织化学法测定过氧化氢酶活性和琥珀酸脱氢酶活性。结果:实验选取健康清洁级SD大鼠24只,全部进入结果分析。①造模后光镜下各组大鼠肌细胞形态学观察结果:假手术对照组肌细胞正常,肌纤维完整;缺血再灌注组肌细胞部分溶解,肌纤维扭曲、断裂;经典缺血预处理组肌细胞点片状坏死,肌纤维基本完整。②各组大鼠血清丙二醛和总超氧化物歧化酶含量的比较:与缺血再灌注组比较,经典缺血预处理组的血清丙二醛含量明显降低[(11.22±0.94),(7.67±1.21)μmol/L,P<0.05],血清总超氧化物歧化酶含量明显升高[(97.97±7.71),(145.74±11.07)Nu/mL,P<0.05]。③各组大鼠骨骼肌过�
AIM: To observe whether there are protective effects of ischemic preconditioning (IP) on ischemic reperfusion injury of skeletal muscle in rats with enzymohistochemical technique.
METHODS: The experiment was conducted in the Laboratory of Pharmacology, Jinzhou Medical College from July to October 2005. Twenty-four SD rats of clean grade aged one month were selected and randomly divided into 3 groups'. Sham-operation group, isehemic-reperfusion group, clssic IP group with 8 rats in each group. ①skeletal muscular ischemic/reperfusion models in isehemic reperfusion group and classic IP group were established. Rats received intraperitoneal anesthesia and then cut open the skins along the crural sheath. Femoral arterioveins were seperated, skins, muscles and nerves of ill lips were cut off with femoral arterioveins remained, blood supply of femoral artery were blocked up with micro vessel clamps to make models of ischemic posterior limbs. ②Mter 60-minute-ischemia, rats in the isehemic reperfusion group were perfused for 20 minntes, Femoral artefioveins of rats in the classic IP group were blocked up by non-trauma vascular clamps, 10-minute short ischemia and 10-minute reperfusion were given; Femoral arterioveins were re-blocked up after 3 circulations, skin-incisions in front of left thigh were sew up; 60 minutes after ischemia, non-trauma vascular damp was taken out by outing out incision in front of the left thigh. Blood flow recovered without thrombogenesis. After arterial blood recovered, affected claws of rats tured from deep purple to pink, and then reperfused for 20 minutes. Rats in the sham-operation group received braid without deligation, femoral arterioveins exposed for 50 minutes. ③ Skeletal myoarchitectoic changes in each group were observed under light microscope, meanwhile, activitise of superoxide dismutase (SOD) in skeletal muscular tissues of rats in each group were measured with xanthine oxidase (XO) method, content of malondialdehyde (MDA) in serum was measured with th
