摘要
目的:探讨HIF-1α减轻瘢痕内缺氧环境的机制。方法:采用免疫组化法检测烧伤后肉芽、不同时期瘢痕和正常皮肤中缺氧诱导因子-1α(hypoxia-induciblefactor1α,HIF-1α)、血管内皮细胞生长因子(vascularendothelialgrowthfactor,VEGF)、血红素氧化酶-1(hemeoxygenase-1,HO-1)的表达,采用权重方法分别对各组瘢痕的HIF-1α、VEGF、HO-1表达结果进行量化,分析各自规律及相互间关系。结果:随瘢痕的成熟,HIF-1α、VEGF、HO-1表达强度均逐渐减弱,相互间具有正相关性(P<0.01)。结论:HIF-1α、VEGF、HO-1在瘢痕成熟过程中起重要作用,HIF-1α可能通过诱导VEGF、HO-1的表达,促进血管生成、成熟,增加组织内供氧,来减轻瘢痕内缺氧环境。
Objective To explore the mechanism of hypoxia inducible factor 1α(HIF-1α) ameliorating the hypoxia in the course of scar maturation. Methods Expression of HIF-1α,vascular endothelial growh factor(VEGF),and heme oxygenase-1 (HO-1) in burn wound granulation,burn scars of different stages,and normal skin were detected by immunohistochemical staining, The expressions were quantified by weight method,and then the relationships between HIF-1α and VEGF,HO-1 were investigated. Results With the scar maturarion, the expression of HIF-1α, VEGF, HO-1 were all gradually decreased .There were both positive relationships between HIF-1α and VEGF (P〈 0.01), between HIF-1α and HO-1 P〈0.01). Conclusion HIF-1α induces VEGF and HO-1 expression, VEGF promotes angiogenesis,and HO-1 advances vessel maturation,which increase offering oxygen,then the hypoxia is gradually ameliorated during the scar maturarion.
