苯那普利和缬沙坦联合应用对肾病大鼠肾脏Ⅳ型胶原和纤维连接蛋白的影响 被引量:10

Effects of the Combination Treatment with Benazepril and Valsartan on Collagen Ⅳand Fibronectin in the Kidney of Nephritic Rats

摘要 目的探讨苯那普利和缬沙坦联合应用对肾病大鼠细胞外基质Ⅳ型胶原和纤维连接蛋白的影响。方法55只雄性SD大鼠,分为对照组、模型组、苯那普利组、缬沙坦组、联合组。后4组在单侧肾切除1周后给予阿霉素5 mg.kg-1尾静脉注射。苯那普利组给予苯那普利6 mg.kg-1.d-1灌胃,缬沙坦组给予缬沙坦20 mg.kg-1.d-1灌胃,联合组给予苯那普利3 mg.kg-1.d-1联合缬沙坦10mg.kg-1.d-1灌胃。12周后用免疫组化检测肾脏Ⅳ型胶原和纤维连接蛋白的表达。结果肾脏Ⅳ型胶原的表达在苯那普利组、缬沙坦组、联合组均较模型组明显减少(P均<0.01),而联合组又低于苯那普利组和缬沙坦组(P均<0.05)。纤维连接蛋白的表达在苯那普利组、缬沙坦组、联合组均较模型组明显减少(P均<0.01),联合组与苯那普利组和缬沙坦组无显著性差异(P>0.05)。结论苯那普利和缬沙坦联合治疗能进一步减少肾脏Ⅳ型胶原的表达,延缓肾小球硬化,联合治疗的疗效优于单独应用。 Objective To explore the effects of the combination treatment with benazepril and valsartan on collagen Ⅳ and fibronectin (FN)in the kidney of nephritic rats. Methods Fifty-five male Sprague-Dawley rats were divided into control group, model group, benazepril treated group, valsartan treated group and co-treated group. The rats from later four groups were cut off one side kidney and injected with adriamycin (5 mg·kg^-1) through tail vein one week later. The rats from later three treated groups were respectively gastric-perfused with benazepril(6mg·kg^-1), valsantan(20 mg·kg^-1), both benazepril (3 mg·kg^-1) and valsantan (10 mg·kg^-1) every day for 12 weeks. Immunhistochernistry was utilized to detect the expression of collagen Ⅳ and FN in the kidney. Results The expression of collagen Ⅳ in benazepril treated group, valsartan treated group and co-treated group was significandy decreased (P〈0.01, respectively), while that in co-treated group was lower than that in benazepril and valsartan group(P〈0.05, respectively). The expression of FN in benazepril treated group and valsartan treated group and co-treated group was significantly reduced, but thexe was no significantly difference among co-treated group and Denazepril and valsartan group. Conclusion The combination of the treatment with Denazepril and valsartan can more reduce the expression of collagen Ⅳ in kidney and postpone development of glonenalosclerosis. The effect of combination treatment with benazepril and valsartan is superior to monotherapy.
