摘要
【目的】观察犬上腔静脉肌袖和右心房游离壁心肌细胞If电流mRNA的表达水平,从分子水平探讨其在生理状态和心律失常发生中的作用。【方法】8只健康杂种犬,取上腔静脉肌袖及右心房游离壁,采用逆转录聚合酶链反应(RTPCR)的方法测定两部位If电流通道亚单位HCN4mRNA的表达水平并进行半定量分析。【结果】If通道亚单位HCN4在上腔静脉肌袖及右心房游离壁中mRNA表达水平分别为(0.207±0.067)和(0.173±0.058),两者差异无显著性(P>0.05)。【结论】不仅右心房中有If通道亚单位HCN4mRNA的表达,上腔静脉肌袖中也有该基因表达,但If通道mRNA在肌袖中的表达量与右心房比并无显著差异,说明生理状态下If电流在上腔静脉肌袖自发活动中可能不起关键作用。
[Objective]To observe the expression of funny current(If)mRNA in cadiocytes of canine superior vena cava and right atrium. [Methods]The cadiocytes of eight dogs in the superior vena eava and right atrium free wall were obtained for measurement of If channel subunit HCN4 using reverse transcription polymerase chain reaction (RT-PCR). [Results]There was no difference in the HCN4 mRNA level between the cadiocytes of superior vena cava and right atrium( P〉0.05). [Conclusion]There is expression of HCN4 in eadiocytes of the superior vena cava and right atrium free wall, but the.difference is not significant. It seems to suggest that If does not play a critical role in the spontaneous activity of superior vena cava cadiocytes.
