摘要 目的研究大鼠前庭组织中上皮钠通道α、β、γ亚基的表达及其意义。方法用兔抗大鼠上皮钠通道α、β和γ亚基的多克隆抗体,采用免疫组化SP法观察大鼠半规管、椭圆囊及球囊组织中α、β、-γENaC的表达模式。结果在大鼠前庭组织中,α、β、γ亚基较一致地分布于椭圆囊斑、球囊斑和壶腹嵴的上皮细胞胞膜和基质细胞,而α亚基在壶腹嵴基质细胞的表达稍弱。结论ENaC的各个亚基分布于前庭的各个区域,形成功能性通道参与内耳内淋巴的调节,从而保持内耳内环境的稳定。 Objective To investigate the distribution and its significance of α、β、and γ subunits of epithelial sodium channel (ENaC) in the rat vestibule, Methods The expression of α-、β-、and γ- ENaC in the rat vestibule was studied immunocytochemically with the specific polyclonal rabbit antibodies against α、β、and γ subunits of rat ENaC. Results All three subunits of ENaC, α、β、and γ , were widely distributed in the vestibule. Strong immunoreactivities of α-、β-、and γ- ENaC were detected in the apical membranes of epithelial cells of the macula utriculi, macula sacculi and ampullary crest. All three subunits were also expressed in the stroma cells though cells subunit immunoreactivity is weaker than the other two subunits, Conclusion Different subunits of ENaC were expressed in various cell regions of the vestibule, which may form the functional channel to regulate the endolymph, thus serve to maintain homeostasis in the inner ear,
