鸭瘟鸭病毒性肝炎二联弱毒疫苗的研究 Ⅱ、稳症性、保存条件及保存期的测定 被引量:2

摘要 本文报道了鸭瘟鸭病毒性肝炎二联弱毒疫苗的稳定性、保存条件和保存期。稳定性的测定是采用了不同保存条件、不同保存期(月)的二联苗,用抗鸭瘟高免血清或抗鸭病毒性肝炎高免血清中和掉二联苗中的相应弱毒后,测定剩下的鸭病毒性肝炎弱毒和鸭瘟弱毒(满度)分别对9日龄鸡压的半数致死置(LD60)和对鸡胚成纤维细胞(CEF)的半数感染量(TCID60)来判定二联苗的稳定性.保存期的测定是采用不同保存条件(℃)不同保存期(月)的二联苗免疫成年鸭,10天后采血测定抗鸭肝炎病毒中和抗体效价及抵抗1万倍最小致死量鸭瘟强毒攻击的能力.试验结果表明:非冻干二联苗子-15—25℃条件下可保存18个月,0℃冻冻结态下可保存1年,4-10℃可保存半年,15-25℃保存10天。疫苗冻干后,-15—25℃条件下可保存20个月,0℃保存问个月,4-10℃保存12个月,15-25℃保存20天. This paper discusses the stability, the condition and period ofbivalent attenuated vaccines against Duck Plague (DP) and Duck Viral Heyatitis(DVH) which should be preserved. The method which was used to detect the stabilityis to use higt-immune serum ageinst DPV or DHV to neutralize the attenuated DPVor attenuated DHV in the vaccines, then to detect the rest attenuated DHV titerswith 9-day chicken embryo(LD60) and the rest attenuated DPV titers with chickenembryo adult-fibre cell8 (CEF, TCID60) separately. The method used to detect theperiod of the vaccines which should be preserved is to use the vaccines preservedin different temperature (C) and periods (monthes) to immune the ducks, then todetect the neutralize antibody titers against DHV in the duck serum and theability with which the immunized ducks can be against the challenge by 10thousand times of M. L. D. virulent DPV after 10 days. The results indieated thatthe non-freeze-dried vaccines in -15--25℃、0℃、4-10℃ and 15-25℃ could bepreserved separately for 18 months, 12 months, 6 months and 10 days, and that thefreeze-dried bivalent attenuated vaccines in -15--25℃、0℃、4-10℃ and 15-25℃could be preserved separately for 20 months, 18 months, 12 months and 20 days
