银杏叶提取物对百草枯诱导PC12细胞凋亡的保护作用 被引量:11

摘要 目的:探讨银杏叶提取物(EGb761)对百草枯(PQ)诱导PC12细胞凋亡的保护作用。方法:4-甲基偶氮唑蓝(MTT)检测PC12细胞活性;流式细胞仪(FCM)测定PC12细胞凋亡百分率;Hoechst33342染色观察细胞凋亡;罗丹明123(Rh123)染色检测细胞线粒体膜电位(MMP)。结果:EGb76120,40μg·mL-1可明显减轻PQ对PC12细胞的细胞毒性,EGb761可使细胞活性增强,降低凋亡百分率,减少细胞凋亡数目,抑制细胞MMP的下降。结论:EGb761对PQ诱导的PC12细胞凋亡具有保护作用,其机制可能与抑制PQ引起的细胞MMP下降有关。 Aim: To explore the protective effect of EGb761 on PQ-induced PC 12 cells cytotoxicity. Methods: The viability was measured by MTT. The apoptosis was monitored by FCM. The morphological change of PC12 cells was observed by Hoechst 33342 staining. Mitochondrial membrane potential (MMP) of PC12 cells was detected by Rh123 staining. Results: EGb761 20, 40 μg-mL^-1 significantly attenuated the cytotoxic effect of PQ. It increased the cell survival as measured with MTT, decreased the apoptosis rate, and inhibited MMP collapse. Conclusion: EGb761 has a possible neuroprotective role in PQ-induced apoptosis of PC12 cells, and the mechanism of the effects of EGb761 may be involved in inhibiting MMP collapse induced by PQ.
