来氟米特与他克莫司合用抑制大鼠心脏移植术后IL-2的产生

摘要 目的评价他克莫司和来氟米特联合应用对大鼠心脏移植排斥反应及IL-2产生的抑制作用。方法采用套管法大鼠心脏颈部异位移植模型,将心脏移植后的大鼠随机分为对照组,他克莫司组,来氟米特组,他克莫司和来氟米特联合用药组,观察各组供心存活时间并检测血清IL-2水平。结果供心存活时间联合用药组明显长于单独用药组(P<0.05)。术后7d,外周血IL-2水平联合用药组明显低于单独用药组(P<0.05)。结论他克莫司和来氟米特联合应用对大鼠心脏移植排斥及IL-2产生的抑制作用优于相同剂量下的单一用药。 [Objective] To evaluate the role of combined use of tacrolimus and Leflunomide(Lef) in prevention of acute allograft rejection and IL-2 production in rats. [Methods] After transplantation, the rats were randomly divided into control group, tacrolimus group, Lef group and tacrolimus+Lef group. Graft survival was observed. 7 days after transplantation, the levels of IL-2 in peripheral blood were examined. [Results] The graft survival of tacrolimus +Lef group was longer than tacrolimus group and Lef group rspectively (P 〈0.05). The levels of IL-2 in the peripheral blood of tacrolimus+Lef group is lower than that of tacrolimus group and Lef group 7 days after transplantation (P 〈0.05). [Conclusion] Combined use of tacrolimus and Lef had better effect in prevention of acute rejection and IL-2 production in allograft rats than monotherapy.
