摘要
报道了λ噬菌体在大肠杆菌Ｗ１４８５中增殖、裂解的条件。用５′－磷酸二酯酶与ＳｅｐｈａｄｅｘＧ－２００凝胶过滤纯化的λＤＮＡ，在琼脂糖凝胶呈一条带。经自行纯化的ＥｃｏＲⅠ限制性内切酶切割后，琼脂糖凝胶电泳上出现６个片段。
Bacteriophage lambda DNA was isolated from Escherichia coli W 1485 transfected with lambda bacteriophage.The purification process involved treatment with 5′ phosphodiesterase and chromatography of Sephadex G 200.By this method,a highly purified λDNA could be prepared.The purfied lambda DNA showed one band on agarose gel electrophoresis,and produced six bands after cleavage by EcoR Ⅰ restriction endonuclease.
