摘要 目的:着重介绍电化学生物传感器在基因诊断、药物分析等方面的医学应用,并展望应用前景和发展方向。资料来源:应用计算机检索Medline2000-01/2005-11的文章,检索词为“ElectrochemicalDNABiosensor”,限定文章语言种类为English;同时检索http://www.wanfangdata.com.cn和http://dx2.cqvip.com2000-01/2005-11的文章,检索词为“生物传感器,医药学,药物分析,基因诊断”,限定文章语言种类为中文。资料选择:纳入标准:①电化学生物传感器的研究。②在医药领域中的应用研究。排除标准:①较陈旧的文献。②综述文献。③重复研究。资料提炼:共收集到86篇与电化学DNA生物传感器相关的文献,其中15篇符合纳入标准,排除的71篇文章系同一类重复性研究和综述文献。资料综合:①DNA传感器的基础研究:电化学生物传感器不仅可以用来识别特定碱基序列的DNA,还可以用来检测DNA的损伤,它能在短时间内对细胞蛋白质、基因及其他与生命过程相关的物质进行信息采集,可以帮助医生从基因层次上了解疾病的发生、发展过程,有助于对疾病的及时诊断和治疗。②临床研究:电化学生物传感器可以广泛地应用于对体液中的微量蛋白、小分子有机物、核酸等多种物质的检测。③生物制药:电化学生物传感器可用于生物制药,原理是采用DNA重组技术或其他生物技术来研制的蛋白质或核酸类药物。④生物传感器的未来:由于生物传感器潜在的巨大优越性,在未来的医学检验中的应用范围必将不断拓展,而广阔的市场需求又将进一步推动生物传感器的迅速发展。结论:随着计算机技术、微制造技术和生物材料学的不断发展,生物传感器技术在医学领域的应用将越来越广泛,它将取代医学上一些传统的检测、化验方法,成为广泛普及的常规分析检测仪器。 OBJECTIVE: To mainly introduce the medical application of DNA biosensors in genetic diagnosis and pharmaceutical analysis, and look into the future of the long view of the application and the developmental direction, DATA SOURCES: An online search of Medline database was undertaken to identify articles published in English between January 2000 and November 2005 by using the keywords of "electrochemical DNA Biosensor". Meanwhile, Chinese articles were also retrieved on the web sites of http://www.wanfangdata.com.cn and http://dx2.cqvip.com, the keywords were "biosensors. medicine, medical analysis, genetic diagnosis" in Chinese. STUDY SELECTION: Inclusive criteria: ①study of electrochemical biosensors;②study about its applications in medicine. Exclusive criteria: dated documents:①) reviews; ② repeated researches. DATA EXTRACTION: Totally 86 papers on the restoration of spinal cord trauma were collected. Among these articles only 15 accorded to the above criteria. The 71 deleted ones were repeated researches and reviews. DATA SYNTHESIS: ① Electrochemical biosensors can not only be used in the recognition of DNA, which contains specific bases sequence but also in detection of the DNA damnifieation. The biosensors can collect data from cell protein, gene and other living matter in a short time. They can help doctors find out the nature of disease and its causes, processes, development, and consequences in gene. They are useful tools to the diagnosis and treatment of disease in time.②Electrochemical biosensors can he widely used in the detection of trace of alhumin organic, substance of smaller molecules, nucleic acid etc in body fluids.③ Electrochemical biosensors are used in biological pharmacy. The mechanism: medicine of protein and nucleic acid, which are produced by DNA reeomposing technology and other biological techniques, is used. ④The biosensor is given great use value and significance in theory.
