摘要 目的观察低分子量肝素对视网膜新生血管形成的抑制作用。方法以高浓度氧诱导C57BL/6J幼鼠建立血管增生性视网膜病变动物模型。治疗组皮下分别注射低分子量肝素3.0U/g(大剂量组)和0.5U/g(小剂量组),每日2次共5d;对照组皮下注射相同体积的平衡盐溶液。用免疫组织化学法对视网膜铺片进行染色,了解视网膜血管改变;用组织切片观察并计数突破视网膜内界膜的血管内皮细胞的细胞核数目,观察低分子量肝素对视网膜新生血管形成的抑制情况。结果治疗组视网膜血管密度减少,突破内界膜的内皮细胞细胞核数目明显减少,与对照组相比差异有显著统计学意义;大剂量组突破内界膜内皮细胞细胞核数目减少,与小剂量组相比差异有显著统计学意义。各组间眼内出血情况无显著统计学差异。结论低分子量肝素可以有效抑制视网膜新生血管形成。 Objective To explore the inhibitory effect of low-molecular-weight heparin on retinal neovascularization. Methods Hyperoxia was induced in ischemic retinopathy model in 35 mice. Eighteen mice received an subcutaneous injections of 3.0 U/g low-molecular-weight heparin twice daily for 5 days as large-dosage group and 17 mice received 0.5 U/g of lowmolecular-weight heparin in the same method as small-dosage group while 15 mice received an subcutaneous injections of same volume of BSS as control. In addition 6 untreated mice served as the normal group. The collagen Ⅳ immunohistochemical staining was used to observe changes of retinal vessels on postnatal day 17. The effect of low-molecular-weight on retinal neovascularization was evaluated by counting the endotheliocyte nuclei of new vessels extending from retina to vitreous in the tissue-slice. Results The reduced density of retinal blood vessels in eyes in the treatment group were found in retinal flatmount. The number of the endotheliocyte nuclei of new vessels extending from retina to vitreous was significantly less in the eyes in the small-dosage group (40. 67±4. 92) and large-dosage group ( 36. 03±4. 40 ) than in control group ( 45.23±6.37 ) respectively ( P 〈 0.01 ). The number of the endotheliocyte nuclei of new blood vessels in the large-dosage group was significantly less than that in small-dosage group (36. 03±4. 40 vs 45.23±6.37 ) ( P 〈 0. 01 ). No statistically significant differences in the number of intraocular hemorrhage were noted between the small-dosage group(5) and large-dosage group(6). Conclusion Subcutaneous injection of low-molecular-weight heparin can inhibit retinal neovascularization.
