摘要
背景与目的探讨食管癌患者外周血淋巴细胞与肿瘤细胞体外药敏试验的相关性。方法与材料采用四甲基偶氮唑蓝[3-(4,5-di methylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide,MTT]法体外药敏试验,检测了32例食管癌患者外周血淋巴细胞和肿瘤细胞对9种临床常用化疗药物的敏感性。结果食管癌患者外周血淋巴细胞和肿瘤细胞对5-氟脲嘧啶(5-fluorouracilum,5-Fu)、顺铂(Diamminedich-loroplatinum,DDP)、卡铂(Carboplatinum,CBP)、氨甲喋呤(Methotrexate,MTX)、环磷酰胺(Cyclophosphamide,CTX)敏感;对阿霉素(Adriamycin,ADM)、长春新碱(Vincristin,VCR)不敏感;对足叶乙甙(Etoposide,Vp16)和吡柔比星(Pirarubicin,THP)两者的敏感性不同。结论食管癌患者肿瘤细胞和外周血淋巴细胞对上述化疗药物敏感率间差异无统计学意义。食管癌外周血淋巴细胞化疗药敏检测对临床选择化疗药物具有重要参考价值。对无法取得肿瘤标本的患者,可用外周血淋巴细胞替代癌细胞进行药敏检测。
BACKGROUND & AIM: To study the correlation between in vitro chemosensitivity of peripheral blood lymphocyte and tumor cells MATERIAL AND METHODS: 3-( 4, 5-dimethyhhiazol-2-yl ) -2, 5-diphenyhetrazolium bromide (MTY) method was used to test the sensitivity of the peripheral blood lymphocytes and the tumor ceils of 32 patients with esophageal carcinoma to 9 kinds of anti-cancer drugs. RESULTS: The peripherals blood lymphocytes and the tumor cells were sensitive to chemotherapeutic drugs of 5-fluorouracil (5-FU), diamminedichloroplatinum(DDP), carboplatinum (CBP), methotrexate (MTX) and cyclophosphamide(CTX), but not sensitive to adriamycin (ADM) , vincristin (VCR) There was a lack of correlation between lymphocytes and tumor cells to VP-16 and THP. CONCLUSION: There were no statistical differences in the degree of sensitivity to the above-mentioned drugs between the peripheral blood lymphocytes and the tumor cells (P 〉 0.05). Peripheral blood lymphocytes may replace tumor cells in clinical chemosensitivity testing.
