摘要
目的研究小鼠肾小体发育过程中细胞增殖与凋亡的变化关系,探讨两者之间的关系及在肾小体发育中的作用。方法采用免疫组织化学方法检测和末端脱氧核苷酸转移酶介导的缺口末端标记法(TUNEL染色法)对发育不同阶段肾小体的细胞增殖和凋亡进行观察。结果肾小体发育早期(Ⅰ、Ⅱ),细胞增殖阳性率明显高于细胞凋亡。肾小体发育中期(Ⅲ、Ⅳ),细胞增殖和细胞凋亡两种行态相伴行。肾小体发育晚期(Ⅴ),细胞增殖和凋亡活动均较弱。结论细胞增殖与凋亡行态在小鼠肾小体发育的整个过程中普遍存在,细胞增殖和细胞凋亡的这种动态平衡关系保证了小鼠肾小体的正常发育成熟。
Objective The cell proliferation and apoptosis of renal corpuscle in normal developing mouse were observed to elucidate the relationship and effect between proliferation and apoptosis in developing renal corpuscle. Methods PCNA immunohistochemisty method, Terminal deoxynucleotidyl transferase mediated dUTP-biotin nick endlabding (TUNELstaining method) were used to observe proliferationand apoptosis within corpuscle at different stages of development. Results In the early stage of developing renal corpuscle (Ⅰ ,Ⅱ ), the positive rate of PCNA was higher than apoptosis. In the medium term of development ( Ⅲ, Ⅳ ), two kinds of behaviors were accompanied. In the later period of development ( Ⅴ ), the activities of proliferation and apoptosis were both weak. Conclusions Proliferation and apoptosis exist universally in the course of developing renal corpuscle. The dynamic relation of proliferation and apoptosis guarantee the regular development of renal corpuscle in mouse.
