Abstract The Wuzhishan micropig, with a body weight of 25-35 kg at maturation, was originally distributed in the Wuzhishan Mountains, Hainan Province,China, but it became extinct there a few years ago. This minipig breed has been rescued by the Institute of Animal Science, Chinese Academy of Agricultural Sciences, where a small population(over 100 individuals) has been established by introducing two sows and one boar from the native haunt to Beijing.Since growth hormone(GH) locus is related to the growth of animals, we examined by Southern blotting with a porcine GH cDNA probe the GH locus of the introduced boar and one of the sows and their progeny of this breed as well as individuals of two other breeds (Large White and Fengjing).It was found that, whatever EcoRI, EcoRV or Pst I was used, some of the Wuzhishan minipigs had a strong hybridization band, whereas it was absent in the tested Large White and Fengjing pigs.Whether this difference is related to differentiation in body size of these breeds needs further study, In addition, DNA fingerprinting with human minisatellite 33.15 revealed that similarity coefficient between the introduced boar and sow of the Wuzhishan minipig was 0.632, and that among the progeny population was 0.738±0.047.This suggests that the Wuzhishan minipig might undergo intensive inbreeding and have high tolerance to inbreeding.Similarity coefficient between the Wuzhishan minipig and Large White pigs and between Wuzhishan minipigs and Fengjing pigs was 0.152±0.058 and 0.138±0.059, respectively, indicating that the Wuzhishan minipighas a genetic background dissimilar to other normal pigs.
