摘要
目的探讨 VIP 族神经肽PACAP 对胆囊运动的影响。方法将豚鼠胆囊制备成1.2mm×10mm 的平滑肌条标本4条,固定在组织浴槽中,并连接等张换能器。以 pH7.4的 Krebs-Ringer缓冲液灌流,温度37℃,并通与95% O_2和5% CO_2。胆囊条对神经肽的效应,用ACh 作标准化处理。结果 VIP 对胆囊条有浓度依赖性的舒张作用;PACAP 则呈现出浓度依赖性收缩反应,PACAP38的作用弱于 PACAP27。结论 PACAP 和 VIP 在对离体胆囊运动的作用方面,呈相反的效应。
AIMS To study the effects of new hypothalamic peptides,PACAP 38 and PACAP 27 on the motility of the gallbladder compared with VIP,using isolated guinea pig gallbladder strips. METHODS The guinea pigs(body weight,440-680 g)were sacrified and four gallbladder muscle strips(1.2 mm×10 mm)were prepared.They were then sus- Pended in an organ bath(4 ml)containing Krebs-Ringer solution(pH 7.4) maintained at 37℃,and aerated with 95% O_2 and 5% CO_2 and were connected to isotonic displacement transducers.The responses of gallbladder to the pep- tides were standardized using acetycholine. RESULTS VIP relaxed gallbladder strips in a concentration-related manner. PACAP 38 and 27,on the other hand,had concentration-related excitatory effects.The potency of PACAP 38 was less than that of PACAP 27. CONCLUSIONS The effect of PACAP on the motility of the guinea pig gallblad- der is opposite to that of VIP.
