摘要
目的探讨骨桥蛋白(OPN)在胶质瘤中的表达及相互之间的关系。方法采用免疫组化染色及逆转录酶聚合酶链反应(RTPCR),免疫印迹法(Westernblot)等方法检测正常脑组织和胶质瘤组织中OPNmRNA及蛋白的表达。结果正常脑组织细胞中无OPN表达,而胶质瘤细胞膜和细胞浆及血管内皮细胞中有OPN表达。随着人脑胶质瘤病理分级的增高,OPN的表达明显升高,在高、低度恶性度肿瘤之间也存在明显差异(P<0.05)。结论脑胶质瘤组织表达OPN并与其恶性程度有关。
Objective To investigate the expression of osteopontin (OPN) in human gliomas and the relationship between them. Methods OPN expression in human normal brain cells and ghoma cells was detected by immunohistochemical staining, semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot method. Results OPN, which was not expressed in normal human brain cells, presented in glioma cell membrane, cytoplasm and blood vessel endothelium. In human brain glioma, mean optical density value for osteopontin increased accordingly with the advancing pathological grades. Significant difference was seen in optical density value between low grade ( grade Ⅰ or Ⅱ ) and high grade ( grade Ⅲ or Ⅳ ) ( P 〈 0. 05 ). Conchtsion The expression of osteopontin is closely related to the malignant degree of glioma,
