甲基强的松龙对大鼠颅脑损伤后ICAM-1和TNF的影响

摘要 目的观察甲基强的松龙对大鼠颅脑损伤后血脑组织中ICAM-1和TNF含量的影响,并探讨其作用机制.方法将55只SD大鼠随机分为3组,采用骨窗形成后硬膜外打击法造成鼠脑挫裂伤模型,正常组5只,麻醉后,只行开颅手术,不作头颅打击;治疗组大鼠致伤后即刻腹腔内注射30mg/kg甲基强的松龙,对照组则即刻腹腔内注射30mg/kg生理盐水.对照组和治疗组大鼠分别在伤后6h、24h、48h、72h、96h时间点断头取脑,对大鼠脑外伤后脑组织中TNF含量进行检测.结果大鼠脑皮质中的ICAM-1和TNF活性在伤后6h后较对照组升高显著(p<0.01),48h达高峰并开始下降,96h降至基础水平.甲基强的松龙治疗组在伤后6h^48h ICAM-1和TNF活性较损伤组明显降低(p<0.05).结论颅脑损伤后,受损脑组织中ICAM-1和TNF含量升高,甲基强的松龙可通过抑制损伤后ICAM-1和TNF活性,起到保护创伤神经元的作用. Objective The experiment was conducted to observe the rule of methylprednisolone according to the change of TNF level in brain trauma tissue of rat. Methods 55 rats were made by frontoparictal bone window plasty with extradural hitting,and were divided to three groups: treated group( treated with methylprednisolone 30mg/kg after hitting), contral group (treated with physiolugic .saline' after hitting )normal group (only expose extradural without hitting and injection). The levels of TNF were measured at different points uf time after hitting. Results ICAM - 1 and TNF level in rat cerebral cortes was higher than that in the contrasting group post Iramnatic 6 hour. and reaed to peak post- traunmtic 48 hour and dropped to the basic level post- traumatic 72 hour. The levels of ICAM- 1 and TNF in Methylprednisolone treatment group was obviously lower than that in the injured group post - traumatic 6 - 48hour. Conclusions Methylprednisotone is effecttive on head injury,
