摘要
目的建立一种简便、可靠的荷人乳腺癌MCF-7细胞移植瘤裸鼠动物模型,以供survivin反义寡核苷酸对乳癌荷瘤裸鼠的抑瘤作用研究。方法常规培养MCF-7人乳腺癌细胞株细胞,注射于30只雌性裸鼠右前肢腋下皮下,以移植后肿瘤体积生长到直径为0.5cm为成瘤,并将这些实验动物分成3组分别以阳离子脂质体、硫化反义寡核苷酸(ASODN)或无义寡核苷酸在不同时间对荷瘤裸鼠的瘤体及瘤周注射3次。观察肿瘤抑制率、细胞凋亡率及细胞凋亡情况。结果成功建立了荷人乳腺癌MCF-7细胞移植瘤裸鼠动物模型,应用Survivin反义核酸技术实验治疗发现ASODN/Lip组抑瘤率达70.1%,细胞凋亡率为38.6%,与其他两组比较有显著性差异(P<0.05)。结论应用裸鼠前肢腋下皮下注射法建立的荷人乳腺癌MCF-7细胞移植动物模型方法简便,成瘤率高,可用于对人乳腺癌反义基因治疗的动物实验研究。
Objective To establish a nude mice transplantation tumor model with human breast cancer for study of gene therapy. Methods MCF - 7 Ceils were grown in monolayer cell culture. 30 nude mice were inoculated with 2 ×10^7 ceils of MCF - 7. These nude mice were divided into three groups: Lipofectin, NODN/Lip and ASODN/Lip. The observation was made by tumor restriction rate and apoptosis rate. Results The subcutaneous tumor model in nude mouse was successfully built. The overall rate of tumor implantation and growth was 100 percent in ASODN/ Lip group; the restriction rate reached 70.10%; the apoptosis rate was 38.6%; and these rates were statistically different from the other two groups (P 〈 0.05). Conclusions This model is highly reproducible and is useful in the further study of breast cancer gene therapy.
