Analysis of the functional expression of P2X7 receptor in leukemia patients

摘要 目的研究P2X7受体在白血病患者中的功能性表达及与白血病治疗的关系。方法采用半定量RT-PCR、流式细胞术和细胞内钙离子测定的方法,从核酸、蛋白和功能水平研究白血病患者骨髓单个核细胞(BMMC)P2X7受体的表达和功能。结果白血病患者和正常人标本中可检出P2X7受体的表达,核酸、蛋白和功能水平结果一致。AML、ALL、CML患者P2X7受体表达阳性率高于正常人;但AML各亚型中P2X7受体阳性强度存在差异,M2组低于正常人,M5,M6组高于正常人;按照P2X7受体阳性强度分组,P2X7受体高表达患者诱导治疗后的完全缓解率显著低于P2X7受体阴性和P2X7受体低表达患者。结论P2X7受体在白血病患者BMMC中有功能性表达,其高表达与患者治疗不敏感相关。 Objective To study the functional expression of P2X7 receptor in leukemia patients and its association with therapy. Methods Semi-quantitative RT-PCR, flow cytometry and intracellular free calcium detection were used to study the expression of P2X7 receptor in BMMC samples at mRNA, protein and function levels. Results P2X7 receptor expression could be detected in BMMC from leukemia patients and normal donor at mRNA, protein and function levels. The positive rates in AML, ALL and CML were higher than that of normal control, whereas the positive intensities varied among AML sub-types. The value was lower in M2 group whereas higher in Ms and M6 groups than normal control. When patients were subgrouped according to the expression of P2X7 receptor, after induction therapy, the complete remission rate in high P2X7 receptor group was lower than that in P2X7 receptor negative group or low P2X7 receptor group. Conclusion P2X7 receptor was functionally expression in BMMC from leukemia patients. The high expression of P2X7 receptor was related to insensitivity to therapy.
