摘要 目的:探讨胰腺组织中蛋白激酶Cα(PKCα)和整合素β1的表达及其临床意义。方法:采用免疫组织化学SP方法,检测56例胰腺癌(高、中分化36例,低分化20例;TNM分期Ⅰ~Ⅱ期24例,Ⅲ~Ⅳ期32例;有淋巴结转移30例,无淋巴结转移26例)及23例正常胰腺组织中PKCα和整合素β1的高表达情况。结果:胰腺癌组织中PKCα和整合素β1高表达率分别为62.50%和71.43%,均高于正常胰腺组织(21.74%,43.48%;P均<0.05);PKCα高表达与胰腺癌分化程度、TNM分期和淋巴结转移有关,整合素β1高表达与胰腺癌TNM分期和淋巴结转移有关。在胰腺癌组织中,PKCα表达与整合素β1呈正相关。结论:PKCα和整合素β1的表达和(或)功能异常可能共同参与胰腺癌的发生发展,联合检测2种蛋白对于预测胰腺癌转移潜能具有一定参考价值。 Aim : To investigate the expression of protein kinase C-α(PKCα) and integrinβ1 in pancreatic carcinoma tissue. Methods : The expression of PKCα and integrinβ1 in 56 cases of pancreatic carcinoma and 23 cases of normal pancreatic tissue were studied using immunohistochemical staining technique. Results:The increased expression rates of PKCαand intergrinβ1 in pancreatic neoplasm tissue were 62.50% , and 71.43% , which were significantly higher than those in normal pancreatic tissue, respectively( 21.74% ,43.48% , P 〈 0.05 ) ; the increased PKCα expression was correlated with differentiation degree, TNM stage, and lymphatic metastasis; the increased integrinβ1 expression was correlated with TNM stage and lymphatic metastasis. In pancreatic carcinomas tissue,the expression of PKCα was positively correlated with that of integrinβ1. Conclusion:PKCα and integrinβ1 may play an important role in the carcinogenesis and tumor progression of pancreatic cancinoma, and they may be useful parameters for predicting tumor invasion and metastasis.
