PKC信号传导对ITP患儿外周血T细胞凋亡及Fas/FasL蛋白表达的影响

摘要 目的探讨蛋白激酶C(PKC)信号传导对ITP患儿外周血T细胞凋亡及其相关蛋白Fas、FasL表达的影响。方法无菌采集ITP患儿及正常健康儿童的外周抗凝血,分离T细胞,并分为三组,第一组为空白对照组,第二组加入PKC的激活剂PMA,第三组加入PMA与抑制剂H-7;在37℃、5%CO2条件下培养24小时,用流式细胞仪检测T细胞的凋亡率及Fas、FasL蛋白的表达。结果加入PMA培养液的正常组,T细胞的凋亡率和Fas、FasL蛋白表达与空白对照比较,差别均无显著性差异(P>0.05);加入PMA培养液的ITP组,T细胞的凋亡率与空白对照比较,差别有统计学意义(P<0.05),与加入PMA培养的正常组比较,差别也有显著性(P<0.05),ITP患儿T细胞Fas的表达与其它各组比较,差别均有统计学意义(P<0.05),而FasL的表达仅与ITP组空白对照相比有统计学意义(P<0.05);同时加入PMA和H-7培养液的ITP组,T细胞的的凋亡率和Fas、FasL蛋白表达与只加入PMA培养液的ITP组比较,差异也有显著性(P<0.05),而与空白对照比较,差别均无显著性(P>0.05)。结论PKC信号传导对ITP患儿T细胞凋亡及其相关蛋白Fas、FasL的表达产生影响,并且可能与ITP的免疫病理机制有关。 Objective To investigate the effect of protein kinase C (PKC) signal transduction on expression of Fas/FasL and apoptosis in peripheral blood T lymphocytes in ITP children. Methods Peripheral blood T lymphocytes from ITP children and normal heathy children were isolated. Then T cells were divided into three groups ( A, B and C ) and cultured for 24h under the condition of 37℃, 5% CO2. Group A is blank, PMA was put into group B and PMA + H - 7 was put in to group C. We detected T cells apoptosic rate and expression of Fas/FasL by FCM. Results T Lymphocyte apoptosic rate:com- pared with blank control, normal group with PMA culture medium was of no significance ( P 〉 0. 05 ) , compared with blank control and normal group with PMA culture medium, ITP group with PMA had signi- ficant difference(P 〈0. 05 ). ITP group with PMA and H -7 culture medium had significant difference(P 〈0. 05 )only compared with ITP group with PMA culture medium. Expression of Fas/FasL on T lympho- cytes:compared with blank control, normal group with PMA culture medium was of no significance (P 〉 0. 05 ) compared with normal groups and the other ITP groups, Fas expression of ITP group with PMA hadsignificant difference(P 〈 0. 05 ), but FasL expression of ITP group with PMA had significant difference (P 〈0. 05) only compared with the blank control in ITP groups. ITP group with PMA and H -7 culture medium had significant difference( P 〈0. 05)only compared with ITP group with PMA culture medium, but had 1.1o significance compared with blank control ( P 〉 0. 05 ). Conclusion PKC signal transduction ways can affect T lymphocytes apoptosis and expression of Fas/FasL in ITP children, besides, it can relate to immunity pathogenesis of ITP.
