漏芦甾酮总提取物改善小鼠记忆障碍的实验研究 被引量:9

摘要 目的研究漏芦甾酮总提取物对小鼠学习记忆功能的影响,并初步探讨其作用机制。方法采用跳台法、避暗法及水迷路法观察漏芦甾酮总提取物对小鼠学习记忆功能的影响;采用震颤强度评分法评价药物作用与中枢胆碱能神经系统的相关性。结果漏芦甾酮总提取物能显著对抗东莨菪碱所致的记忆获得障碍及环己酰亚胺所致的记忆巩固障碍;显著对抗东莨菪碱所致的小鼠空间辩别障碍;增强氧化震颤素所致小鼠震颤的强度。结论漏芦甾酮总提取物有改善小鼠学习记忆障碍的作用,该作用可能与其增强中枢胆碱能神经系统功能有关。 Objective To study the effects of total-sterone extracts of Rhaponticum uniflorum on learning and memory and to probe into the therapeutic mechanism preliminarily. Methods The methods of multiple training active avoidance response that include step-down test, step-through test and water maze for mice were used to observe the effect on learning and memory. At the same time, the correlation of effects of the extracts with the central cholinergic system was measured by tremor strength grading method. Results Total sterone-extracts of Rhaponticum uniflorum could markedly counteract memory acquisition impairment induced by scopolamine(SCPL) and memory consolidation impairment induced by cycloheximide, significantly counteract spatial discrimination impairment induced by SCPL in mice, and enhance the strength of tremor induced by oxotremorine in mice. Conclusion Total sterone-extracts of Rhaponticum uniflorum can improve the ability of learning and memory. The mechanism may be related with the improvement of the function of the central cholinergic system.
