摘要 目的研究不同剂量的能量可控陡脉冲对体外培养的人脐静脉血管内皮细胞生物学性状的影响。方法对人脐静脉血管内皮细胞施以不同剂量的陡脉冲,采用四甲基偶氮唑盐(MTT)比色法及细胞增殖曲线观察细胞的损伤效应,流式细胞技术分析作用前后细胞周期的情况。结果①MTT试验证明能量可控陡脉冲对人脐静脉内皮细胞的作用存在剂量-效应关系,其杀伤率随剂量的增加而增加,揭示高剂量的能量可控陡脉冲可导致血管内皮细胞急性损伤效应;②细胞增殖曲线表明随能量可控陡脉冲剂量的增加,对细胞增殖能力抑制越明显;③流式细胞分析结果表明,能量可控陡脉冲处理前后,细胞周期变化不明显。结论能量可控陡脉冲对血管内皮细胞有明显的杀伤和抑制作用。 Objective To investigate the effects of energy controllable steep pulse on the biological properties of human umbilical vein endothelial cells (HUVECs) in vitro. Methods After action of steep pulses of 10 V and 80 V, the acute and delayed effects on human umbilical vein endothelial cells were investigated by measuring the cell growth curves and by MTT assay, and cell cycle changes were detected by flow cytometry (FCM), manner. Results The cell MTT assay demonstrated the effects of steep pulses on HUVECs were in a dose-dependent growth curve showed that the higher dose mainly resulted in acute effects, but there were no changes in cell cycle. Conclusion It shows that energy controllable steep pulse can change the feature of human umbilical vein endothelial cells and induce the apoptosis of HUVEC.
