摘要
目的诱导建立小鼠淋巴管内皮细胞肿瘤模型。方法8周龄C57BL/6小鼠16只,腹腔注射弗氏不完全佐剂与PBS(1∶1)乳悬液,间隔15天1次,共注射2次。分4批处死小鼠后,对诱导出的小鼠腹腔新生物进行病理组织学观察,采用免疫组织化学方法检测淋巴管内皮细胞标志性抗原VEGF-C和Flt-4。另以4只小鼠同样方法注射等量PBS液作为对照。结果13只(81.3%)实验小鼠膈肌腹腔面、肝脏表面及侧腹壁腹腔面可见边界清楚的散在分布白色肿瘤样组织,光学显微镜和透射电镜观察发现为由内皮细胞构成的多管腔囊性结构,并表达淋巴管内皮细胞标志性抗原VEGF-C和Flt-4。对照小鼠中未见新生物。结论小鼠腹腔注射弗氏不完全佐剂可稳定诱导小鼠腹腔淋巴管内皮细胞肿瘤模型。
Objective To establish an animal model of benign lymphangiomas in nice.Methods Sixteen C57BL/6 mice accepted twice intraperitoneal injection with incomplete Freund, s adjuvant with 15 days interval. Neoplasms induced were studied histopathologically. The expressions of VEGF-C and Fit-4 were examined by immunohlstochemical method in neoplasms specimen. Results White solid tumor masses were induced in peritoneal cavity in 13 of 16 mice(81.3 % ). The histological study of tumors was lymphangioma,and tumor cells expressed VEGF-C and Fit-4 positively. Conclusion Intraperitoneal injection with incomplete Freund,s adjuvant is a good method to induce benign lymphangiomas in mice.
