摘要 研究SephadexG-25,G-50,G-100,G-200和G-200(超细)等型号的葡聚糖凝胶系统,对溶解在1.0%SDS-2Me水溶液中的网纹藤壶Balanusreticulatus固态胶蛋白组分的分离制备效果。结果表明,SephadexG-200和G-200(超细)两种葡聚糖凝胶层析柱(直径1.7cm,长30cm),用0.05mol·L-1Tris-HCl-1%SDS-2Me溶液洗脱时,均可将改蛋白组分按其分子量的大小进行分离,且以后者的分高效果更好。 Because of the possible application as an underwater adhesive and its importance in antifouling fields, great attention has been gived to the physicochemical properties and polymerization mechanisms of barnacle cement. In this paper, the separation of barnacle cement components by the Sephadex G-25, G-50, G-100, G-200, as well as G-200 (superfine ) polydextran gels for preparative purposes was studied. 50 mg solified cement of Balanus reticulatus was dissolved in 5 ml 1%SDS-2Me solution, and a 17mm X 300mm gel filtration column was used. The eluent was 0. 05mol· L-1 Tris-HCl-1% SDS-2Me.Due to the absorption of mercaptoethanol near 280 nm which disturbed the detection of proteins, tile fractions were detected by the 5%-20% gradient SDS-polyacrylamide gel electrophoresis after elution from the molecular sieving column as above. The resultsobtained showed that the components (subunits) of barnacle cement were separated on the basis of their molecular weights by means of the Sephadex G-200 or G-200 (superfine),and if hat the latter was better. This separation method will help us to obtain sufficient samples for furthering the study on the relationship, particularly the cross-link form between those components and understanding the attachment mechanisms of barnacles. In addition, some problems that occurred in the experiments were also discussed.
