摘要
目的探讨鱼油联合肿瘤坏死因子α(TNFα)真核表达载体对乳腺癌移植瘤生长的影响。方法将人乳腺癌细胞(MCF-7)接种到BABL/c裸鼠右侧腋部皮下建立移植瘤模型,将动物随机分为4组:对照组、鱼油组、TNFα组和鱼油+TNFα组。实验结束时观察移植瘤的大小,流式细胞仪分析移植瘤的细胞周期相分布,RT-PCR分析移植瘤组织中tmTNFαmRNA水平。结果单纯tmTNFα真核表达载体转染对移植瘤生长无明显影响。鱼油组和鱼油+TNFα组与对照组比较肿瘤的重量显著降低,其中鱼油+TNFα组肿瘤最小。鱼油处理后,移植瘤细胞G0/G1所占百分比增加,S期细胞减少。鱼油+TNFα组tmTNFαmRNA水平比对照组高,而鱼油组和TNFα组tmTNFαmR-NA表达水平与对照组差异无统计学意义。结论鱼油在体内能诱导外源性tmTNFα在移植瘤组织中高效表达,并增强鱼油对乳腺癌移植瘤的抑制作用。
Objective To explore the effect of fish oil and transmembrane TNFα eukaryotic expression vector on breast cancer xenograft growth in nude mice. Methods MCF-7 ceils were transplanted subcutareously to BALB/c nude mice to produce tumor. The nude mice with xenograft were randomly divided into four groups: control group, fish oil treated group, TNFα transfacted group and fish oil + TNFα transfacted group. The xenograft tumor growth in mice was observed, The ceil cycle was measured by using flow cytometer. The expression of transmembrane TNFα mRNA was determined by RT-PCR. Results Fish oil significantly inhibited the tumor growth in vivo. The cell cycle analysis showed that fish oil blocked the tumor cells at the G1/G0 phage so as to reduce the ratio of S phage. These changes were more obviously in fish oil + TNFα transfacted group than those of fish oil treated group. The expression of transmembrane TNFα mRNA was increased in fish oil + TNFα transfacted group. Conclusion Fish oil could induce expression of transmembrane TNFα in xenograft tumor trans-facted with pPPRE-tk-tmTNFα, and this could promote the anti-tumor activity of fish oil in vivo.
