摘要
以莪术粗提物为原料,应用高速逆流色谱(HSCCC)技术,通过对制备分离方法和溶剂系统的筛选,采用溶剂系统正己烷-乙酸乙酯-乙醇-水和正己烷-乙醚-乙醇-水,转速880 r/m in,流速1.5 mL/m in,进样量30 m g,检测波长254 nm,记录仪衰减为200,分离得到姜黄素。通过外标物的对照,1H-NMR进行结构鉴定,HPLC进行纯度分析,确定得到的物质为姜黄素,纯度为94.06%。
To establish a method for the isolation and purification of curcumin from the crude exact of Rhizoma curcumae by high-speed counter current chromatography, study was focused on the selection of solvent system and extraction method. The solvent system used is hexane-aether-ethanol-water and hexane-ethyl acetate-ethanol-water, the rotate speed is 880 r/min, flow rate is 1.5 mL/min, sample injected is 30 mg, ),λ=254 nm. Through the analysis of 1H-NMR and HPLC, and with the comparison of the standard substance, the curcumin with the purity of 94.06 % was obtained.
