反相高效液相色谱法检测红霉素发酵过程中S-腺苷甲硫氨酸

摘要 建立用反相高效液相色谱(RP-HPLC)检测红霉素发酵液中S-腺苷甲硫氨酸(SAM)的方法,观察其在发酵过程中的变化。样品经离心、超声等处理,进行RP-HPLC分析,梯度洗脱。实验结果说明,该方法可以使SAM与发酵液中其它复杂组分得到较好的分离。红霉素发酵过程中SAM量随时间呈曲线变化,高峰值出现在96h左右。 Reverse phase high-performance liquid chromatography (RP-HPLC) was applied to detersyl-l-methionine (SAM) in erythromycin fermentation course. Fermentation broth samples HPI.C analysis after centrifugation and sonication and other processes, then they were eluted under gradient condition (solvent A ,0.25mol/L ammonium acetate, pH6.0, solvent B, 100% methanol, 20min gradient, 0-40% B), flow rate was 1.0ml/min, detection was carried at 254nm. The results showed that SAM and other complex compounds in the fermentation broth could be separated fairly well by this method. SAM concentration was changed in a curve mode during the erythromycin fermentation with a peak at 96h.
