缺血再灌注大鼠视网膜细胞线粒体钙含量与组织型纤溶酶原激活物活性的相关性 被引量:1

摘要 目的观察缺血再灌注大鼠视网膜细胞线粒体钙含量与组织型纤溶酶原激活物(tissue-type plasminogen acti-vator,TPA)活性的变化,探讨两者之间的相关性。方法采用提高眼压法造成视网膜缺血后,恢复眼压形成血流再灌注。实验分正常对照组和缺血1 h再灌注1 h组、缺血1 h再灌注2 h组、缺血2 h再灌注1 h组、缺血2 h再灌注2 h组四个实验组,每组各取10例测定视网膜细胞线粒体钙浓度与TPA活性的变化,并进行相关性分析。结果缺血再灌注后,大鼠视网膜细胞线粒体钙含量与TPA活性随缺血和再灌注时间的延长而升高(P<0.01),两者之间呈显著相关性(r=0.524,P<0.01)。结论缺血再灌注可引起视网膜细胞线粒体钙含量的升高,从而导致视网膜组织TPA活性的增高,引起视网膜组织结构和功能的损伤。 Objective To observe the changes SD rat retinas in the concentration of mitochondrial calcium and the activity of the tisse-type plasminogen activator (TPA) in ischemia and reperfusion and to study the correlation between them. Methods Intraocular pressure(IOP) was raised to ocelude retinal blood vessels and create retinal ischemia IOP was then reduced to a normal level to allow reperfusion. The experimrimental design included a control group and 4 ischemia-reperfusion groups. Retinal tissaes of 10 rats from each group were rneasured for mitochondrial calcium concentration and TPA activity and the correlation between them was analyzed. Results Both mitochondrial calcium concentration and TPA activity in the control group were lower than that in the 4 experimental groups ( P 〈 0.01). When the duration of ischmiareperfusion was longer, mitochondrial calcium concentration and TPA activity were higher. There was a positive correlation between them (r =0.524, P 〈 0.01).Conclusion. Ischemia and reperfusion can stinmlate a rkqe in mitochondrial calcium concentration in the rat retina, which leads to an increase in TPA activity and damage to retinal structure and function.
